摘要
目的:探究LINC01287在甲状腺癌中的表达及对细胞增殖、侵袭的影响。方法:收集16例甲状腺癌患者的癌组织及其癌旁组织(距癌组织3 cm)标本,体外培养人正常甲状腺上皮细胞TEC61及人甲状腺癌细胞(BCPAP、TPC-1、FTC-133、8505C、SW1736),qRT-PCR检测甲状腺癌组织和细胞中LINC01287表达情况。利用细胞转染技术分别将LINC01287-shRNA(sh-LINC01287)及其阴性对照(sh-NC)转染至FTC-133细胞中(sh-LINC01287组、sh-NC组),同时设置正常培养的对照(Control)组。MTT实验、克隆形成实验检测各组FTC-133细胞增殖情况;Transwell实验检测各组FTC-133细胞侵袭情况;Western Blot实验检测各组FTC-133细胞增殖、侵袭相关蛋白(细胞周期蛋白D1,Cyclin D1;神经型钙黏附蛋白,N-cadherin;基质金属蛋白酶9,MMP-9)表达情况。结果:LINC01287在甲状腺癌组织中的表达水平高于癌旁组织(P<0.05);LINC01287在BCPAP、TPC-1、FTC-133、8505C、SW1736细胞中的表达水平均高于TEC61细胞,且在FTC-133细胞中最高(P<0.05),故选取FTC-133细胞进行后续实验;与Control组和sh-NC组比较,sh-LINC01287组FTC-133细胞的增殖率降低,克隆形成数、侵袭数及Cyclin D1、N-cadherin、MMP-9蛋白表达均减少(P<0.05)。结论:LINC01287在甲状腺癌中高表达,敲低LINC01287表达可抑制FTC-133细胞增殖和侵袭,LINC01287可能是甲状腺癌潜在的治疗靶点或预后生物标志物。
Objective:To explore the expression of LINC01287 in thyroid cancer and its influ ences on cell proliferation and invasion.Methods:The cancer tissues and their adjacent tissues(3 cm from the cancer tissues)of 16 patients with thyroid cancer were collected,Human normal thyroid epithelial cells TEC61 and human thyroid carcinoma cells(BCPAP,TPC-1,FTC-133,8505C,SW1736)were cultured in vitro,and the expression of LINC01287 in thyroid cancer tissues and cells was detected by qRT-PCR.The LINC01287-shRNA(sh-LINC01287)and its negative control(sh-NC)were transfected into FTC-133 cells(sh-LINC01287 group,sh-NC group)using cell transfection technology,meantime,the normal culture control group was set up.The proliferation of FTC-133 cells in each group was detected by MTT assay and clone formation assay;the invasion of FTC-133 cells in each group was measured by Transwell assay;and the expression of proliferation and invasion of FTC-133 cells related proteins(Cyclin D1;N-cadherin;matrix metalloproteinase 9,MMP9)in each group was measured by Western Blot assay.Results:The expression level of LINC01287 in thyroid cancer tissues was higher than that in adjacent tissues(P<0.05);the expression level of LINC01287 in BCPAP,TPC-1,FTC-133,8505C and SW1736 cells was higher than that in TEC61 cells,and was the highest in FTC-133 cells(P<0.05),so FTC-133 cells were selected for follow-up experiments;Compared with the Control group and sh-NC group,the proliferation rate,the number of colonies formed,the number of invasive cells,and the protein expressions of Cyclin D1,N-cadherin,and MMP9 of FTC-133 cells in sh-LINC01287 group were all decreased(P<0.05).Conclusion:LINC01287 is highly expressed in thyroid cancer,knockdown of LINC01287 can inhibit the proliferation and invasion of FTC-133 cells,and LINC01287 may be a potential therapeutic target or prognostic biomarker for thyroid cancer.
