摘要
目的观察火针对膝骨性关节炎(KOA)模型大鼠血清肿瘤坏死因子-α(TNF-α)及软骨组织p38的影响,探讨火针对KOA起效的机制。方法抽签法将15只SD大鼠均分至3组(5只/组),分别标为空白组、模型组与火针组。除空白组大鼠外,其余动物均建立KOA模型。火针组采用毫火针点刺治疗,隔日1次,治疗2周。ELISA法分析大鼠血清TNF-α含量,膝关节软骨组织内p38 mRNA表达水平用实时荧光定量PCR法(qPCR)检测。结果模型组大鼠血清TNF-α含量相较于空白组升高明显(P<0.01),膝关节软骨组织p38 mRNA相对表达量亦有增加,差异具统计学意义(P<0.05);与模型组相比,火针组血清TNF-α明显降低,差异有显著统计学意义(P<0.01),p38 mRNA表达减少,差异同具有统计学意义(P<0.05)。结论火针可通过降低外周血内TNF-α的含量,抑制软骨组织内p38的基因表达对膝骨性关节炎起治疗作用。
Objective To observe the effects of fire acupuncture on serum TNF-αand cartilage p38 in rats with knee osteoarthritis and explore the possible mechanism of fire acupuncture for treating knee osteoarthritis.Methods Fifteen SD rats were randomly divided into blank group,model group and fire acupuncture group,with five rats in each group.Except for the blank group,the other rats were established with knee osteoarthritis models.The fire acupuncture group was treated with fire acupuncture once every other day for two weeks.The serum TNF-αcontent of the rats were analyzed by Elisa and the p38 mRNA expressions in the knee cartilage tissue were detected by quantitative real-time PCR(qPCR).Results Compared with the blank group,the serum TNF-αcontent and the mRNA expression of p38 in knee cartilage of the rats of the model group significantly increased and the differences were statistical significant(P<0.01,P<0.05).Compared with the model group,the serum TNF-αof the fire acupuncture group significantly reduced(P<0.01).The mRNA expression of p38 of the fire acupuncture group was lower than that of the model group and the difference was also statistically significant(P<0.05).Conclusion Fire acupuncture has a therapeutic effect on knee osteoarthritis by reducing the content of TNF-αin peripheral blood and inhibiting the gene expression of p38 in cartilage tissue.
