摘要
卵巢癌是妇科三大恶性肿瘤之一,病死率最高、预后最差。根据组织学特点,卵巢癌又可分为多种类型,其中上皮性卵巢癌(EOC)是最常见的卵巢癌类型,复发率高,且易发生腹腔转移,影响患者预后。EOC腹腔转移的机制研究已经取得一定进展,研究认为通过一系列复杂的病理过程,癌细胞可以通过悬浮形成单个或多个球体形式,利用肌球蛋白产生的力清除覆盖腹腔表面的间皮细胞,实现癌细胞局部侵犯盆腔和腹部器官,但对其具体潜在分子机制尚未达成共识。同时,部分研究者深入研究发现EOC细胞系球囊形成实验过程中腺苷酸活化蛋白激酶(AMPK)及自噬通量LC3表达升高,相关自噬抑制剂增加了癌细胞对化疗药物的敏感性,可能成为潜在的治疗靶点。本文以AMPK介导的自噬为介入点对EOC腹腔转移的研究进展进行综述,并对该研究未来的发展方向进行展望。
Ovarian cancer is one of the three major malignant tumors in gynecology,with the highest mortality and the worst prognosis.According to histological characteristics,ovarian cancer can be divided into various types,in which epithelial ovarian cancer(EOC)is the commonest type of ovarian cancer,with high relapse rate and easy peritoneal metastasis,exerting impacts on the prognosis of patients.The research on the mechanism of EOC peritoneal metastasis has made certain progress.It is believed in the researches that through a series of complicated pathological processes,cancer cells can be suspended to form single or multiple spheres,and the mesothelial cells covering the abdominal surface can be removed by the force generated by myoglobulin,so that cancer cells can locally invade pelvic cavity and abdominal organs.However,no consensus has been reached on its specific potential molecular mechanism.At the same time,some researchers found that the expression of AMP-activated protein kinase(AMPK)and autophagy flux LC3 increased during the balloon formation experiment of the EOC cell line,and related autophagy inhibitors increased the sensitivity of cancer cells to chemotherapy drugs,which may become potential therapeutic targets.In this paper,AMPK-mediated autophagy is taken as the intervention point to review the research progress of EOC peritoneal metastasis,and the future development direction of this research is prospected.
