阿苯达唑—葡聚糖纳米颗粒体外抗多房棘球蚴原头节的效果评价

摘要 目的 通过观察阿苯达唑-葡聚糖纳米颗粒(ABZ-GPs)和阿苯达唑(ABZ)对多房棘球蚴原头节的杀伤作用,评价ABZ-GPs对多房棘球蚴病的疗效,为葡聚糖颗粒作为抗包虫病药物载体奠定基础。方法 设置ABZ、葡聚糖纳米颗粒(GPs)组、ABZ-GPs组和对照组,各用药组以终浓度50μg/mL加入原头节培养基,每天用0.4%的台盼蓝染液染色以评价原头节存活情况,并使用扫描电镜(SEM)和透射电镜(TEM)观察原头节超微结构的变化。结果 成功构建了ABZ-GPs,原头节与ABZ-GPs和ABZ共培养后,外翻型原头节明显增加,体表失去正常结构。培养第7 d开始至第10 d, ABZ-GPs组原头节存活率一直低于ABZ组,差异有统计学意义(t=3.091,P<0.05)。超微结构显示对照组和GPs组原头节吸盘清晰可见,体表微绒毛蓬松,角质层细胞形态均匀饱满;ABZ组原头节褶皱,吸盘变形,小钩排列紊乱,角质层见絮状空洞的分裂区域;ABZ-GPs组原头节明显褶皱,微绒毛消失,吸盘破坏,顶突小钩脱落,角质层广泛损伤,呈条状残留。结论 ABZ-GPs在体外对多房棘球蚴的杀伤作用优于ABZ原药。 This study was aimed at evaluating the efficacy of ABZ-GPs on alveolar echinococcosis,to lay a foundation for glucan particles as anti-echinococcosis drug carrier.The killing effects of ABZ-GPs and ABZ on the Echinococcus multilocularis protoscoleces were observed.An ABZ group,GPs group,ABZ-GPS group and control group were analyzed.Protoscoleces culture medium containing different treatments was added at a final concentration of 50μg/mL,and 0.4%trypan blue staining solution was used to evaluate daily survival.Scanning electron microscopy(SEM)and transmission electron microscopy(TEM)were used to observe the ultrastructural changes in protoscoleces segments.ABZ-GPs were successfully constructed.After co-culture of ABZ-GPs and ABZ with protoscoleces,the valgus protoscoleces segments significantly increased,and the body surface lost its normal structure.From day 7 to day 10 of culture,the survival rate of protoscoleces in ABZ-GPs group was always lower than that in ABZ group,and the difference was statistically significant((t=3.091,P<0.05)).Ultrastructure showed that the protoscoleces suckers of the control group and GPs group were clearly visible,the microvilli on the body surface were fluffy,and the cuticle cells were uniform and full;in the ABZ group,the protoscoleces was folded,the suckers were deformed,the small hooks were arranged disordered,and the cuticle showed the division of floccus void;in the ABZ-GPS group,the protoscoleces was obviously folded,the microvilli disappeared,the sucker was destroyed,the apical hooks fell off,and the cuticle was extensively damaged,showing a strip-like residue.Comprehensively,the solubility of ABZ-GPs in vitro and its killing effect against Echinococcus multilocularis were superior to those of ABZ.
