摘要
目的探讨血清中miR-143-5p和miR-34a-5p在多囊卵巢综合征患者中的表达水平及与PCOS患者临床病理特征的关系,评估miR-143-5p和miR-34a-5p作为诊断PCOS的生物标志物潜能。方法选取2019年1月至2021年6月在南京市第一医院收治确诊为PCOS的患者40例(PCOS组)和同期50例体检正常的非PCOS女性(对照组),分析两组研究对象的生殖激素及胰岛素相关指标,采用qRT-PCR检测miR-143-5p及miR-34a-5p表达水平,ROC曲线分析miR-143-5p及miR-34a-5p对PCOS的诊断价值。结果血清中基础LH值PCOS组为(7.88±1.21)mIU/mL,对照组为(5.46±1.21)mIU/mL,两组相比差异具有统计学意义(P<0.001);空腹胰岛素PCOS组为(9.37±2.88)mIU/L,对照组为(8.04±1.98)mIU/L,组间相比差异具有统计学意义(P=0.036);PCOS组胰岛素抵抗指数和睾酮显著高于对照组,两组间均有统计学差异(P<0.05)。PCOS组血清中miR-143-5p及miR-34a-5p表达量较对照组显著升高,差异有统计学意义(P<0.01)。miR-143-5p作为诊断PCOS患者血清学标志物,ROC曲线下面积为0.836,95%CI为0.732~0.884;miR-34a-5p作为诊断PCOS患者血清学标志物,ROC曲线下面积为0.771,95%CI为0.648~0.838。结论miR-143-5p及miR-34a-5p在PCOS患者血清中高表达,可作为PCOS的血清分子诊断方法。
Objective To investigate the expression level of miR-143-5p and miR-34a-5p in serum in patients with polycystic ovary syndrome and the relationship between them and the clinicopathological characteristics of PCOS patients,and to evaluate the potential of miR-143-5p and miR-34a-5p as biomarkers of PCOS.Methods A total of 40 patients diagnosed with PCOS(PCOS group)and 50 healthy non-PCOS females(control group)who were admitted to Nanjing first Hospital from January 2019 to June 2021.The reproductive hormones and insulin related indexes of the two groups were analyzed.The expression levels of miR-143-5p and miR-34a-5p were detected by qRT-PCR,and the diagnostic value of miR-143-5p and miR-34a-5p for PCOS was analyzed by ROC curve.Results The basal LH value in serum was(7.88±1.21)mIU/mL in PCOS group and(5.46±1.21)mIU/mL in control group.Fasting insulin PCOS group was(9.37±2.88)mIU/L and control group was(8.04±1.98)mIU/L.Insulin resistance index and testosterone in PCOS group were significantly higher than those in control group,and there were statistical differences between the two groups(P<0.05).The expression levels of miR-143-5p and miR-34a-5p in blood of PCOS group were significantly higher than those of control group,with statistical significance(P<0.01).miR-143-5p was used as a serological marker for the diagnosis of PCOS patients.The area under ROC was 0.836,95%CI:0.732-0.884.miR-34a-5p as a serological marker for the diagnosis of PCOS patients,had an area under ROC was 0.771,95%CI of 0.648-0.838.Conclusion miR-143-5p and miR-34a-5p are highly expressed in the serum of PCOS patients,which can be used as serum molecular diagnostic methods for the diagnosis of PCOS.
