死亡细胞释放信号对肿瘤进展的影响及中医药干预 被引量:2

摘要 细胞存在多种多样的死亡途径,根据发生机制的不同,可分为凋亡、坏死性凋亡、焦亡、铁死亡、自噬依赖性死亡等。事实上,死亡细胞不是等待移除的惰性细胞,其可通过释放可溶性细胞因子及包含丰富生物学成分的囊泡等作为“再见”信号,进一步影响周围组织及细胞的生物学功能。值得注意的是,近几十年来细胞死亡领域相关研究多聚焦于细胞死亡的内在调控过程,而对细胞死亡后释放信号的多样性,以及所释放信号如何调控肿瘤的发生发展关注较少。一方面,死亡细胞释放信号可调控肿瘤炎性微环境形成、代谢、耐药、转移等各种生物学过程。此外,死亡细胞释放信号可作为第二信使,调控肿瘤微环境的免疫应答反应活性。合理利用多种多样的细胞死亡途径并采用药理学手段调节细胞死亡释放信号,有望为肿瘤的防治提供新的治疗方向及策略。中医药在调节细胞死亡信号进而抑制肿瘤发生发展领域具有传统优势。拟综述主要探讨细胞死亡途径的释放信号对肿瘤生物学行为的影响,以及中医药调控细胞死亡途径进而抗肿瘤的作用,以期为中医药抗肿瘤临床应用提供理论依据。 Cells die in various ways,such as apoptosis,necroptosis,pyroptosis,ferroptosis,autophagy-dependent cell death and so on.In fact,dead cells are not inert cells waiting to be removed.They can release the soluble cytokines and various vesi⁃cles that contained different biological components as the“goodbye”signals,and therefore affecting the biological functions of the surrounding tissues and cells.Notably,the researches on cell death in recent decades have mainly focused on the internal modula⁃tion of the cell death process,while the knowledge about the diversity of dead cell-derived signals as well as their biological effects on cancer occurrence and development is very limited.On one hand,dead cell-derived signals can regulate various bio⁃logical processes of cancer,such as inflammatory microenvironment formation,metabolism,drug resistance and metastasis.Addi⁃tionally,dead cell-derived signals can also act as the second messenger to regulate the immune response activity within the tumor microenvironment.Pharmacological regulation of dead cell-derived signals and various cell death pathways is expected to provide potential intervention strategies for cancer prevention and treatment.Traditional Chinese medicine(TCM)is very effective in regulating cell death signals-mediated tumorigenesis and cancer progression.This review intends to summarize the biological effects of dead cell-derived signals on the biological behaviours of tumors as well as the modulatory effect of TCM on cell death signals.This review may provide a novel theoretical basis for the anti-tumor application of TCM in the clinic.
