摘要
单核细胞增生李斯特菌(Listeria monocytogenes,Lm)是一种重要的食源性人兽共患病原菌,可通过食物链感染人类,严重危害人体健康。因此快速检测食物样品中Lm对于李斯特菌病的防控尤为必要。细胞壁表面蛋白ActA是Lm重要的毒力因子,在前期制备的ActA单克隆抗体的基础上,将其与磁珠进行偶联,并结合李斯特菌显色培养基快速检测Lm。结果表明:抗体最适偶联量为100μg,最佳偶联时间为4 h,细菌最佳富集时间为15 min。在此基础上建立了免疫磁珠快速检测技术,具有特异性强、灵敏度高等优点。综上,该技术可高效检测出食品中的Lm,对预防和控制李斯特菌病的发生具有重要意义。
Listeria monocytogenes(Lm)is an important zoonotic foodborne pathogen,which can infect human beings through the food chain and seriously endanger human health.Rapid detection of Lm in food is necessary for the prevention and control of listeriosis.The cell surface protein ActA is an important virulence factor of Lm.Here,the prepared ActA monoclonal antibody was coupled with magnetic beads,combining with Listeria chromogenic medium,then the method was utilized to rapid detection of Lm.The results showed that the optimal conjugation amount of antibody was 1ooμg,the best conjugation time was 4 h,and the optimum enrichment time of bacteria was 15 min,on this basis,the immunomagnetic bead detection technique was established,which had the advantages of high specificity and sensitivity.Detection of Lm in food with high efficiency is of great significance to prevent and control the occurrence of listeriosis.
