摘要
试验旨在研究不同剂量的黑沙蒿水提物对奶牛体外瘤胃发酵功能的影响。试验采用单因素完全随机试验设计,以3头体况相近的健康荷斯坦奶牛为瘤胃液供体动物,在全混合日粮中(TMR)中分别添加0(对照组)、0.025%(H1组)、0.050%(H2组)、0.075%(H3组)、0.100%(H4组)的黑沙蒿水提物,培养24 h,测定瘤胃发酵参数和发酵气体成分。结果表明,与对照组相比,不同剂量黑沙蒿添加组奶牛瘤胃液中pH值、NH_3-N含量均显著降低(P<0.05),菌体蛋白(BCP)含量显著升高(P<0.05)。H2组、H3组、H4组奶牛瘤胃液NH_3-N含量均显著低于对照组和H1组(P<0.05)。H3组、H4组奶牛瘤胃液原虫数显著低于对照组(P<0.05)。H1组、H2组和H3组奶牛瘤胃液二氧化碳含量显著高于对照组(P<0.05),氢气含量显著低于对照组(P<0.05)。与对照组相比,H2组、H3组奶牛瘤胃总挥发性脂肪酸、乙酸、丙酸、戊酸浓度显著增加(P<0.05);H3组奶牛瘤胃异戊酸浓度显著高于其他各组(P<0.05)。研究表明,添加黑沙蒿水提物会降低瘤胃pH值,抑制原虫的繁殖,促进瘤胃发酵,以0.050%~0.075%剂量组的效果较好,尤以0.075%剂量最佳。
The experiment was to investigate the effect of different doses of Artemisia annua water extract on rumen fermentation of dairy cows in vitro.Three healthy Holstein dairy cows with similar body conditions were selected as rumen fluid donors,and 0(control group),0.025%(H1 group),0.050%(H2 group),0.075%(H3 group)and 0.100%(H4 group)of Artemisia annua water extracts were added to the total mixed diet(TMR),respectively.Rumen fermentation parameters and fermentation gas components were determined after 24 h culture.The results showed that compared with control group,the pH value and NH3-N concentration in rumen fluid of cows in different dosages of Artemisia annua supplemental groups were significantly decreased(P<0.05),while the content of BCP was significantly increased(P<0.05).NH3-N content in rumen fluid of dairy cows in H2 group,H3 group and H4 group was significantly lower than that in control group and H1 group(P<0.05).The number of rumen fluid protozoa in H3 group and H4 group was significantly lower than that in control group(P<0.05).The content of carbon dioxide in rumen fluid of cows in H1 group,H2 group and H3 group was significantly higher than that in control group(P<0.05),and the content of hydrogen in rumen fluid of cows in H1 group,H2 group and H3 group was significantly lower than that in control group(P<0.05).Compared with control group,the concentrations of total volatile fatty acids,acetic acid,propionic acid and valeric acid of rumen fluid of cows in H2 group and H3 group increased significantly(P<0.05),and the concentration of isovaleric acid in rumen fluid of cows in the H3 group was significantly higher than that in other groups(P<0.05).The experiment indicates that adding water extract of Artemisia annua water extrac can reduce rumen pH value,inhibit protozoa reproduction and promote rumen fermentation,and the effect of 0.050%~0.075%dose group is better,especially 0.075%dose.
