野黄芩苷对血管性痴呆大鼠学习能力和Trx-1、TrxR-1的影响

Effects of wild baicalin on learning ability,Trx-1 and TrxR-1 in rats with vascular dementia

摘要 [目的]探讨野黄芩苷对血管性痴呆大鼠学习记忆功能的影响及机制。[方法]选取SD大鼠60只,随机分为:假手术组、模型组、阳性对照组(地塞米松6 mg/kg)、野黄芩苷低、中、高剂量组(100 mg/kg、200 mg/kg、400 mg/kg);构建血管性痴呆大鼠模型,并灌胃相应药物治疗。采用mNSS表对大鼠神经功能评分,Morris水迷宫实验检测大鼠学习和记忆能力,RT-qPCR检测Trx-1、TrxR-1 mRNA,Western Bloting检测Trx-1、TrxR-1蛋白。[结果]与假手术组比较,模型组大鼠mNSS评分、逃逸时间显著增加(P<0.05),停留时间、穿越次数、Trx-1、TrxR-1 mRNA和蛋白水平显著降低(P<0.05);在给予野黄芩苷和地塞米松干预后,与模型组比较,阳性对照组和野黄芩苷低、中、高剂量组mNSS、逃逸时间均显著降低,停留时间、穿越次数、Trx-1、TrxR-1 mRNA和蛋白水平显著升高,且呈剂量依赖性(P<0.05);阳性对照组与野黄芩苷高剂量组各指标比较无显著差异(P>0.05)。[结论]野黄芩苷可有效活化海马组织中Trx-1、TrxR-1表达水平,从而改善血管性痴呆大鼠的学习记忆功能。 [Objective]To investigate the effect of Baicalin improving learning and memory ability in rats with vascular dementia and its effect on expression of Trx-I and TrxR-I in hippocampus.[Method]Sixty SD rats were selected and randomly divided into:sham operation group,model group,positive control group(dexamethasone,6 mg/kg),low-dose scutellarin(100 mg/kg),medium-dose scutellarin(200 mg/kg),high-dose scutellaringroup(400 mg/kg).A rat model of vascular dementia was constructed,and the corresponding drugs were administered by gavage.The neurological function of the rats was scored by mNSS table,the learning and memory abilities of the rats were detected by the Morris water maze test,and RT-qPCR was used to detect Trx-1 and TrxR-I mRNA,Western Bloting used to detect the Trx-1,TrxR-I protein in rat brain tissue.[Result]Compared with the sham-operated group,the mNSS score and escape time in the model group were significantly increased(P<0.05).Time,number of crossings,Trx-1,TrxR-1 mRNA and protein levels were decreased(P<0.05).After the intervention of baicalin and dexamethasone,compared with the model group,the mNSS scores of the positive control group and baicalin low,medium and high dose groups,and escape time were all significant decrease,dwell time,crossing times,Trx-1,TrxR-1 mRNA and protein levels were increased in a dose-dependent manner(P<0.05).There was no significant difference in each index between the positive control group and the high-dose scutellarin group(P>0.05).[Conclusion]Baicalin can effectively improve the learning and memory function of vascular dementia rats,and its mechanism may be related to reducing oxidative stress by activating the expression of Trx-I and TrxR-I in the body.
