东寨港红树林湿地沉积物抗生素抗性基因的分布特征及其影响因素

Distribution Characteristics of Antibiotic Resistance Genes in Mangrove Wetland Sediments of Dongzhai Port and the Influencing Factors

摘要 红树林是生长于热带与亚热带潮间带的一类常绿灌木及小乔木群落,其生境极易受污染影响,是抗生素抗性基因(ARGs)的储存库。以海南省东寨港红树林湿地演丰西河为研究区,通过采集研究区沉积物样品,利用宏基因组测序技术,分析研究区沉积物中ARGs的分布特征及其影响因素,并对沉积物中ARGs与环境因子、微生物群落结构的相关性进行分析。结果表明:研究区沉积物中共检测出212种ARGs亚型,所属12种抗生素抗性类型以利福霉素、氨基糖苷、大环内酯、肽类和磺胺类为主,主要的抗性机制为抗生素靶点改变与靶点置换;研究区不同区域沉积物中优势ARGs基本一致,但不同深度沉积物中的优势ARGs略有差别;Pearson相关分析结果显示,研究区内沉积物中有25种ARGs与环境因子具有显著的相关关系(p<0.05),且相对丰度前20名的ARGs除Paer_CpxR(多药耐药抗性基因)外,均与一个或多个细菌属具有不同程度的相关性(p<0.05)。通过全面评估东寨港红树林湿地沉积物中ARGs污染,可为红树林湿地环境中ARGs传播的生态风险分析与ARGs污染治理提供科学依据。 Mangroves are a group of evergreen shrubs and small tree communities that grow in tropical and subtropical intertidal zones.Their habitats are highly vulnerable to pollution and are reservoirs of antibiotic resistance genes(ARGs).In this paper,taking the Yanfenxi River in the mangrove wetland Dongzhai Port,Hainan Province as the study area,we investigated the distribution characteristics of ARGs in sediment and their influencing factors using macrogenome sequencing technology,and analysed the correlation between ARGs in sediment,environmental factors,and microbial community structure.The result shows that there are 212 ARG subtypes in sediment of the study area,belonging to 12 antibiotic resistance types,mainly rifamycin,aminoglycosides,macrolides,peptides and sulfonamides,and the main resistance mechanisms are antibiotic target alteration and target replacement.Pearson correlation analysis shows that 25 ARGs in the study area correlate with environmental factors(p0.05) significantly,and all the top 20 ARGs in abundance except Paer_CpxR(multidrug resistance genes)correlate with one or more bacterial genera to different degrees(p0.05).Through a comprehensive review of ARGs contamination in the mangrove wetland sediments of Dongzhai Harbor,this study provides basis for the risk of ARGs transmission and the mechanism of ARGs contamination in mangrove environments.
