摘要
目的:了解IL-17A基因rs8193037及rs8193038位点多态性与川崎病的相关性。方法:纳入川崎病患儿及健康儿童各100例,应用聚合酶链式反应(polymerase chain reaction,PCR)和DNA直接测序法检测2组IL-17A基因rs8193037及rs8193038的单核苷酸基因多态性,比较2组目标等位基因频率的分布情况。结果:对于IL-17A基因rs8193037位点,川崎病组中GG、GA、AA基因型频率为70%、27%、3%,G、A等位基因分布频率分别为83.5%、16.5%;健康对照组GG、GA、AA基因型频率为72%、26%、2%,G、A等位基因分布频率分别为85%、15%,2组差异无统计学意义(χ^(2)=0.17,P>0.05)。对于IL-17A基因rs8193038位点:川崎病组GG、GA、AA基因型频率结果为72%、25%、3%,G、A等位基因分布频率为84.1%、15.9%;健康对照组中GG、GA、AA基因型频率结果为72%、26%、2%,G、A等位基因分布频率为84.6%、15.4%,2组差异无统计学意义(χ^(2)=0.01,P>0.05)。结论:IL-17A基因中rs8193037及rs8193038位点多态性与川崎病无明显关联性。
Objective:To investigate the association of the polymorphisms of IL-17A gene rs8193037 and rs8193038 with susceptibility of Kawasaki disease.Methods:A total of 100 children with Kawasaki disease and 100 healthy children were enrolled in this experiment.The polymorphisms at positions IL-17A gene rs8193037 and rs8193038 in the 2 groups were detected by polymerase chain reaction(PCR)and direct DNA sequencing in order to compare the distribution of target allele frequencies in the 2 groups.Results:The distribution frequencies of GG,GA,and AA genotypes of IL-17A rs8193037 in the Kawasaki disease were 70%,27%,and 3%,and the distribution frequencies of G and A alleles were 83.5%and 16.5%,respectively.In the healthy children group,the distribution frequencies of GG,GA,AA genotypes were 72%,26%,and 2%,and the distribution frequencies of G and A alleles were 85%and 15%,respectively.There was no significant statistical difference between the 2 groups(χ^(2)=0.17,P>0.05).Moreover,the distribution frequencies of GG,GA,and AA genotypes of IL-17A rs8193037 in the Kawasaki disease were 72%,25%,and 3%,and the distribution frequencies of G and A alleles were and 84.1%and 15.9%,respectively.In the healthy children group,the distribution frequencies of GG,GA,and AA genotypes were 72%,26%,and 2%,and the distribution frequencies of G and A alleles were 84.6%and 15.4%,respectively.There was no significant statistical difference between the 2 groups(χ^(2)=0.01,P>0.05).Conclusion:No significant correlation was found between the polymorphisms of loci rs8193037 and rs8193038 in IL-17A gene and Kawasaki disease.
