莱菔硫烷对D-半乳糖致衰老模型小鼠海马组织氧化应激水平的影响

The Effect of Sulforaphane on Oxidative Stress in Hippocampus of Aging Mouse Induced by D-glactose

摘要 目的探讨莱菔硫烷(sulforaphane,SFN)对衰老模型小鼠海马组织氧化应激水平的影响。方法将24周龄健康小鼠按体质量随机分为对照组、模型组、干预组,每组10只,雌雄各半。模型组和干预组小鼠皮下注射200 mg·kg^(-1) D-半乳糖,每日1次;干预组小鼠给予25 mg·kg^(-1) SFN灌胃,每日1次,模型组小鼠灌胃等量蒸馏水;对照组小鼠皮下注射等量生理盐水,灌胃等量蒸馏水。干预至48周龄后,在末次给药2 h后取脑并称重。左侧海马组织用于检测超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)和总抗氧化能力(T-AOC)活力,右侧海马组织用于检测mRNA与蛋白水平。结果与对照组相比,模型组小鼠海马组织GSH-Px、CAT、T-AOC活性、Nrf2 mRNA水平均降低,MDA含量升高(P均<0.05);与模型组相比,干预组小鼠海马组织GSH-Px、SOD、CAT、T-AOC活性、Nrf2 mRNA和蛋白水平均升高,MDA含量降低(P均<0.05)。结论SFN可增强衰老模型小鼠海马组织抗氧化酶类活性及抗氧化能力,减少其氧化损伤。 Objective To investigate effects of sulforaphane on oxidative stress in hippocampus of aging mice.Methods 24 weeks’old mice were assigned to control group,aging model group and SFN intervention group(n=10,equal numbers of male and female mice).Aging model group and intervention group were subcutaneously injected with 200 mg·kg^(-1) D-galactose once a day,and mice in SFN intervention group were treated with 25 mg·kg^(-1) SFN once a day,the mice in the model group were given with equivalent distilled water.The control group was fed with distilled water and the same amount of solvent in the same way.Intervention to 48 weeks of age,and 2 h after the last dose,the brain was isolated and weighed.The left hippocampus was used to detect the activity of superoxide dismutase(SOD),malondialdehyde(MDA),glutathione peroxidase(GSH-Px),catalase(CAT)and total antioxidant capacity(T-AOC).The right hippocampus was used to detect mRNA levels and protein levels.Results Compared with the control group,the activities of GSH-Px,CAT,T-AOC and Nrf2 mRNA in hippocampus of the model group were decreased,and the content of MDA in hippocampus was increased significantly(P all<0.05).Compared with the aging model group,the activities of GSH-Px,SOD,CAT,T-AOC,Nrf2 mRNA and protein levels in hippocampus of the intervention group were increased,and the content of MDA in hippocampus was decreased significantly(P all<0.05).Conclusion SFN could enhance the activities of antioxidant enzymes and the antioxidant capacity,and reduce the oxidative damage in hippocampus of aging mice.
