摘要
目的研究护巢饮对早发性卵巢功能不全大鼠氧化应激及磷脂酰肌醇3-激酶/蛋白激酶B/核因子E2相关因子2/血红素加氧酶-1(PI3K/Akt/Nrf2/HO-1)通路的调节作用。方法取12只SD大鼠作为正常组,取60只SD大鼠采用雷公藤多苷灌胃法建立早发性卵巢功能不全模型。将造模成功大鼠随机分为模型组、护巢饮低剂量组、护巢饮中剂量组、护巢饮高剂量组及戊酸雌二醇组,每组12只。护巢饮低、中、高剂量组大鼠分别给予5 g/kg、10 g/kg、20 g/kg的护巢饮灌胃,戊酸雌二醇组大鼠给予0.1 mg/kg的戊酸雌二醇灌胃,均1次/d,连续15 d。ELISA法测定大鼠血清促卵泡激素(FSH)、雌二醇(E 2)、抗缪勒管激素(AMH)、抑制素B(INH-B)、促黄体生成素(LH)水平及卵巢组织中活性氧(ROS)、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、丙二醛(MDA)含量,称量卵巢湿重、子宫湿重,计算卵巢指数、子宫指数,HE染色法观察大鼠卵巢组织病理学形态,qRT-PCR法测定卵巢组织中Nrf2、HO-1 mRNA表达量,Western blot法测定卵巢组织中p-PI3K/PI3K、p-Akt/Akt、Nrf2、HO-1蛋白表达情况。结果与正常组比较,模型组大鼠血清FSH、LH水平及卵巢组织中ROS、MDA含量均明显升高(P均<0.05),血清E 2、AMH、INH-B水平及卵巢组织中CAT、SOD含量和卵巢湿重、卵巢指数、子宫湿重、子宫指数均明显降低(P均<0.05),闭锁卵泡数量明显增多(P<0.05),初级卵泡、窦前卵泡、窦状卵泡数量均明显减少(P均<0.05),卵巢组织中Nrf2、HO-1 mRNA表达量和p-PI3K、p-Akt、Nrf2、HO-1蛋白表达量均明显降低(P均<0.05);与模型组比较,护巢饮各组及戊酸雌二醇组血清FSH、LH水平及卵巢组织中ROS、MDA含量均明显降低(P均<0.05),血清E 2、AMH、INH-B水平及卵巢组织中CAT、SOD含量和卵巢湿重、卵巢指数、子宫湿重、子宫指数均明显升高(P均<0.05),闭锁卵泡数量明显减少(P均<0.05),初级卵泡、窦前卵泡、窦状卵泡数量�
Objective It is to study the effect of Huchaoyin on oxidative stress and phosphatidylinositol 3-hydroxy kinase(PI3K)/protein kinase B(Akt)/nuclear factor E 2 related factor 2(Nrf2)/heme oxygenase-1(HO-1)pathway in rats with premature ovarian insufficiency.Methods Twelve SD rats were taken as the normal group,and 60 SD rats were taken to establish premature ovarian insufficiency models by gavage with tretinoin.The successfully modeled rats were randomly divided into model group,low-dose Huchaoyin group,middle-dose Huchaoyin group,high-dose Huchaoyin group and estradiol valerate group,with 12 rats in each group.The rats in the low,medium and high dose groups of Huchaoyin were respectively given Huchaoyin 5 g/kg,10 g/kg and 20 g/kg by gavage,and the rats in the estradiol valerate group were given estradiol valerate 0.1 mg/kg by gavage,all administered once/d,continuously treated for 15 d.The serum levels of follicle stimulating hormone(FSH),estradiol(E 2),antimüllerian hormone(AMH),inhibin B(INH-B),luteinizing hormone(LH),and the contents of reactive oxygen species(ROS),catalase(CAT),superoxide dismutase(SOD),and malondialdehyde(MDA)in ovarian tissues of the rats were determined by ELISA,the ovarian wet weights and uterine wet weights were weighed,the ovarian index and uterine index were calculated,the histopathological morphology of rat ovarian tissues was observed by HE staining,the expression of Nrf2 and HO-1 mRNA in ovarian tissues were detected by qRT-PCR,and the protein expression of p-PI3K/PI3K,p-Akt/Akt,Nrf2 and HO-1 in ovarian tissues were detected by Western blot.Results Compared with the normal control group,the levels of serum FSH and LH and the contents of ROS and MDA in ovarian tissues of the rats in the model group were significantly increased(all P<0.05),the levels of serum E 2,AMH and INH-B and the contents of CAT and SOD in ovarian tissues and ovarian wet weight,ovarian index,uterine wet weight and uterine index were significantly decreased(all P<0.05),the number of atretic follicles was significa
