摘要
目的:探讨莪连颗粒对胃癌种植瘤裸鼠肿瘤生长及维甲酸相关孤核受体γt(retinoid-related orphan nuclear receptorγt,RORγt)表达的影响。方法:构建裸鼠人胃癌SGC-7901种植瘤模型,实验组予莪连颗粒灌胃,对照组予生理盐水灌胃。灌胃28天后处死小鼠并测定种植瘤体积及瘤重,计算抑瘤率。采用Western-Blot法检测肿瘤组织中RORγt表达。结果:实验组小鼠瘤重及肿瘤体积均低于对照组,抑瘤率为53.3%;实验组肿瘤组织RORγt表达较对照组减少。结论:莪连颗粒可抑制胃癌肿瘤生长,该作用可能与其抑制RORγt表达,减轻由辅助性T细胞17介导的免疫调节有关。
Objective:To discuss the influence of Elian granule on tumor growth and the expression of RORγt in nude mice with transplanted gastric cancer.Methods:After constructing SGC-7901 implant tumor model of human gastric cancer in nude mice,the experiment group was drenched with Elian granules,and the control group with normal saline.After 28 days of gavage,to detect the tumor volume and tumor weight after the mice were sacrificed,to calculate tumor inhibition rate.Western-Blot was adopted to measure the expressions of RORγt in tumor tissue.Results:The experiment group was lower than the control group in tumor weight and tumor volume,the tumor inhibition rate was 53.3%;the expressions of RORγt in tumor tissue of the experiment group decreased compared with the control group.Conclusion:Elian granules could inhibit the growth of gastric cancer,and its effects might be related to that it could inhibit the expressions of RORγt and relieve the immunoregulation mediated by T helper cell 17(Th17).
