低共熔溶剂提取橄榄多酚及其抗氧化活性评价 被引量:4

Optimization of Deep Eutectic Solvent Extraction Process and Evaluation of Antioxidant Activity of Olive Polyphenols

摘要 探究超声辅助低共熔溶剂法提取橄榄多酚的最佳提取工艺,并考察评价橄榄多酚的抗氧化活性。以橄榄多酚得率为评价指标,在低共熔溶剂的种类、摩尔比、含水量、液料比、超声时间和超声温度等单因素考察基础上,以响应面法进一步优化提取工艺。通过1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)清除试验,以抗坏血酸为阳性对照,与醇提法比较,评价经优化提取的橄榄多酚的抗氧化能力。超声辅助低共熔溶剂法的最佳提取工艺:以含水量为30%的氯化胆碱-乙二醇(1∶4)为提取溶剂,在液料比42∶1 mL/g、超声温度39℃、超声时间25 min条件下,橄榄多酚的得率为7.46%±0.06%。DES法提取的橄榄多酚浓度为14.84μg/mL,清除DPPH的抑制率可高达95.70%,半数抑制浓度(IC_(50))为3.05μg/mL;醇提法提取橄榄多酚IC_(50)为3.07μg/mL;抗坏血酸IC_(50)为6.45μg/mL;橄榄多酚抗氧化活性明显高于抗坏血酸。超声辅助低共熔溶剂法为橄榄多酚的提取提供了一种新的高效提取法,且能保持橄榄多酚良好的抗氧化活性,也为其抗氧化活性功能产品的开发奠定了基础。 To study the optimum extraction process of olive polyphenols by ultrasonic assisted deep eutectic solvent method,and to investigate and evaluate the antioxidant activity of olive polyphenols,the extraction yield of olive polyphenols was taken as the evaluation index,and the response surface test was used to optimize the extraction process on the basis of single factor experiments of deep eutectic solvent type,mole ratio,water content,liquid-to-material ratio,ultrasonic time and ultrasonic temperature.Compared with alcohol extraction method,the antioxidant capacity of the optimized extracted olive polyphenols was evaluated by a 1,1-xylene-2-diphenyl-2-picrylhydrazyl,DPPH) clearance test using ascorbic acid as a positive control.The optimal extraction conditions of ultrasonic assisted deep eutectic solvent extraction process were as follows:choline chloride-glycol(molar ratio 1∶4) with water content was 30% as the extraction agent,liquid-to-material ratio was 42∶1 mL/g,ultrasonic temperature was 39 ℃,ultrasonic time was 25 min,and the extraction yield of olive polyphenols was 7.46%±0.06%.The concentration of olive polyphenols extracted by the DES method was 14.84 μg/mL,the inhibition rate of the DPPH clearance could be as high as 95.70%,and half of the inhibitory concentration(IC_(50)) was 3.05 μg/mL.The IC_(50) of olive polyphenols extracted by Alcohol extraction method was 3.07 μg/mL.The IC_(50) of ascorbic acid was 6.45 μg/mL.The antioxidant activity of olive polyphenols was significantly higher than that of ascorbic acid.Ultrasonic assisted deep eutectic solvent method provides a new and highly efficient extraction method for olive polyphenols extraction,and can maintain the good antioxidant activity of olive polyphenols,which also lays the foundation for the development of its functional products with antioxidant activity.
