摘要
通过单因素试验来筛选碳源、氮源和无机盐的种类,通过响应面试验优化榛蘑液体菌种培养基配方,并研究菌丝体多糖体外抗氧化活性。结果表明:当以蔗糖作为碳源、蛋白胨作为氮源和磷酸二氢钾作为无机盐时,菌丝体多糖含量和生物量最高。榛蘑液体菌种培养基最优配方为蔗糖添加量33.0 g/L、蛋白胨添加量21.6 g/L、磷酸二氢钾添加量1.7 g/L,在此条件下菌丝体多糖含量达到8.91 g/100 g。热水浸提法得到的菌丝体多糖具有清除DPPH·、ABTS^(+)·、·OH的能力,说明榛蘑菌丝体多糖具有一定的抗氧化活性。
The types of carbon source,nitrogen source and inorganic salt were screened by single factor test.The formula of liquid culture medium of Armillaria mellea was optimized by response surface test,and the in vitro antioxidant activity of mycelium polysaccharide was studied.The results showed that the polysaccharide content and biomass of mycelium were the highest when sucrose was used as carbon source,peptone as nitrogen source and potassium dihydrogen phosphate as inorganic salt.The optimal formula of liquid culture medium of Armillaria melea was sucrose supplemental level of 33.0 g/L,peptone supplemental level of 21.6 g/L,potassium dihydrogen phosphate supplemental level of 1.7 g/L,under these conditions,mycelium polysaccharide content reached 8.91 g/100 g.The mycelium polysaccharide obtained by hot water extraction had the ability to clear DPPH·,ABTS^(+).and·OH,which indicated that the mycelium polysaccharide had certain antioxidant activity.
