封闭式电喷雾萃取电离装置的研制及性能测试

摘要 电喷雾萃取电离(EESI)技术具有无需复杂的样品预处理、可直接电离样品中目标分子的特点,广泛用于分析复杂基体样品尤其是气溶胶样品。然而,现有的EESI装置为敞开式结构,实验过程中容易受周围环境因素的影响,特别是气流扰动以及外源气溶胶的干扰。基于此,本工作研制了一种封闭式EESI装置,并在实验室环境条件下采用组氨酸、利血平、L-甲硫氨酰-L-精氨酰-L-苯丙氨酰-L-丙氨酸乙酸盐(MRFA)3种标准溶液测试封闭式与敞开式EESI-MS的性能。结果表明,封闭式EESI装置的日内精密度和灵敏度略优于敞开式EESI装置。在不同外界空气流速下,使用利血平溶液测试上述2种EESI-MS的性能,随着外界空气流速增大,敞开式EESI-MS装置的日内精密度逐渐下降(RSD从3.4%上升到5.6%),封闭式EESI-MS装置的日内精密度良好(RSD从2.4%上升到3.6%),表明封闭式EESI装置具有较强的抗气流干扰能力。采用封闭式EESI装置连续5天分析检测不同浓度的葡萄糖气溶胶,日内精密度(RSD<3.5%)和日间精密度(RSD<8.8%)均良好,表明封闭式EESI装置具有长期工作稳定性。分别使用封闭式与敞开式EESI-MS连续5次分析检测5名志愿者呼出气体样品中的葡萄糖,封闭式EESI-MS装置的日内精密度(RSD<3.7%)明显优于敞开式EESI-MS装置(RSD<8.1%),表明封闭式EESI装置用于检测实际样品具有更好的稳定性。 Extractive electrospray ionization(EESI)technology has the advantages of directly ionizing the target molecules in the samples without complex sample pretreatments,and it is widely used for analysis of complex matrix samples,especially for aerosol samples.However,the current EESI device with open structure is easily affected by the surrounding environmental factors,especially susceptible to the airflow disturbance and exogenous aerosols.To solve this problem,an enclosed-structure EESI device was developed in this work.Three standard solutions of histidine,reserpine and MRFA were used to test the performances of the enclosed and open EESI-MS under laboratory conditions,respectively.The results showed that intra-day precision and sensitivity of enclosed EESI device are slightly better than that of open EESI device.Then,the performances of the two EESI-MS devices were tested with reserpine solution under different external air flow rates.As the external air flow rate increased,the intra-day precision of the open EESI device gradually decreased(RSD increased from 3.4%to 5.6%),and the intra-day precision of the enclosed EESI device remained good(RSD increased from 2.4%to 3.6%),indicating that the enclosed EESI device has a strong ability to resist airflow disturbance.The glucose aerosols of different concentrations were used to test the enclosed EESI-MS for 5 consecutive days.The results showed that intra-day precision(RSD<3.5%)and inter-day precision(RSD<8.8%)are good,indicating that the enclosed EESI device has a long-term working stability.The performances of enclosed and open EESI-MS were evaluated by detecting glucose from the exhaled breath of 5 volunteers within 5 consecutive times,respectively.The intra-day precision of the enclosed EESI device(RSD<3.7%)was significantly better than that of the open EESI device(RSD<8.1%),indicating that the enclosed EESI device has a better stability in real sample detection.
