摘要
目的探讨冠状动脉粥样硬化性心脏病(CAD)患者血浆中长链非编码RNA H19(LncRNA H19)、微小RNA 130a(miRNA 130a)表达水平与冠状动脉狭窄程度的相关性。方法选择湖北医药学院附属国药东风总医院心血管内科2021年7月到2021年10月住院治疗并行冠状动脉造影(CAG)的患者92例,根据CAG的结果分为CAD组(61例)和对照组(31例),通过实时荧光定量PCR(Real-time qPCR)测定各组中LncRNA H19和miRNA 130a的表达水平;CAD组采用Gensini积分来评估冠状动脉狭窄程度,分为低分组(<40分)、高分组(≥40分),比较两组血浆中的LncRNA H19和miRNA 130a的表达量,采用SPSS 25.0统计学软件进行统计分析。结果①CAD组血浆中LncRNA H19表达水平高于对照组,miRNA 130a表达水平低于对照组(P<0.05)。②低分组血浆中LncRNA H19表达水平低于高分组(P<0.05);而低分组血浆中miRNA 130a表达水平高于高分组(P<0.05)。③采用Spearman检验分析,CAD患者血浆中的LncRNA H19表达水平与Gensini评分呈正相关(r=0.481),miRNA 130a的表达水平与Gensini评分呈负相关(r=-0.386)。④LncRNA H19、miRNA 130a以及二者联合诊断CAD的受试者工作特征(ROC)曲线下面积(AUC)分别为0.685、0.804、0.821;LncRNA H19、miRNA 130a表达水平以及两者联合诊断冠状动脉狭窄程度的AUC分别为0.777、0.723、0.836。结论血浆中LncRNA H19和miRNA 130a表达水平可能成为预测冠状动脉狭窄程度的有效指标。
【Objective】To investigate the correlation between the expression levels of long non-coding RNA H19(LncRNA H19)and microRNA 130a(miRNA 130a)in the plasma of patients with coronary atherosclerotic heart disease(CAD)and the degree of coronary stenosis.【Methods】A total of 92 patients who underwent coronary angiography(CAG)in the Department of Cardiovascular Medicine in our hospital from July 2021 to October 2021 were selected.According to the results of CAG,they were divided into CAD group(61 cases)and control group(31 cases).The expression levels of LncRNA H19 and miRNA 130a in each group were determined by real-time quantitative PCR(real-time qPCR);the CAD group was assessed by Gensini score to evaluate the degree of coronary stenosis,and divided into low group(<40 points)and high group(≥40 points),the expression levels of LncRNA H19 and miRNA 130a in the plasma of the two groups were compared,and SPSS 25.0 statistical software was used for statistical analysis.【Results】(1)The expression level of LncRNA H19 in the plasma of the CAD group was higher than that of the control group,and the expression level of miRNA 130a was lower than that of the control group,and the differences were statistically significant(P<0.05).(2)The expression level of LncRNA H19 in the plasma of the low score group was lower than that of the high score group(P<0.05);while the expression level of miRNA 130a in the plasma of the low score group was higher than that of the high score group(P<0.05).(3)Using Spearman test analysis,the expression level of LncRNA H19 in plasma of patients with coronary heart disease was positively correlated with Gensini score(r=0.481),and the expression level of miRNA 130a was negatively correlated with Gensini score(r=-0.386).(4)The area under the ROC curve(AUC)of LncRNA H19,miRNA 130a and their combined diagnosis of CAD were 0.685,0.804 and 0.821,respectively;The AUC of the expression levels of LncRNA H19,miRNA 130 and their combined diagnosis of coronary artery stenosis were 0.777,0.723,0.836,resp
