重复经颅磁刺激对小鼠缺血性脑损伤修复及DJ-1表达的影响

摘要 目的 探讨重复经颅磁刺激(repetitive transcranial magnetic stimulation,rTMS)通过调控DJ-1的表达,使小鼠缺血性脑损伤后的神经功能缺损得到恢复的机制。方法 选取8~12周健康雄性C57BL/6J小鼠30只,随机分为3组:假手术组(Sham组)、缺血再灌注组(MCAO组)和治疗组(rTMS组),每组10只。MCAO组使用线栓法制备大脑中动脉闭塞(middle cerebral artery occlusion,MCAO)小鼠模型,Sham组小鼠处理同MCAO组但不插入线栓。Sham组和rTMS组给予rTMS治疗,MCAO组不予治疗。造模后1 h拔出线栓进行再灌注,再灌注24 h后开始给予rTMS治疗,频率为1 Hz,每次持续25 s,每日5次,连续治疗7 d。在治疗前后对各组小鼠进行神经功能缺损评分;使用TTC染色确定各组小鼠脑组织梗死灶面积;并采用免疫组化技术检测梗死灶周围区域神经元中DJ-1表达变化;使用免疫印迹技术检测DJ-1的蛋白水平表达变化。结果 与Sham组(0.50±0.53)比较,MCAO组(2.80±0.63)和rTMS组(2.10±0.32)的神经功能缺损评分显著增高(P<0.05);与MCAO组比较,rTMS组的神经功能缺损评分显著降低(P<0.05)。与MCAO组比较,rTMS组的梗死灶面积明显减小,DJ-1表达显著提高(P<0.01)。结论 rTMS可有效促进缺血性脑损伤的小鼠神经功能恢复,并显著减少梗死灶面积,可能分子机制是通过上调神经DJ-1蛋白表达干预损伤预后。 Objective To investigate the effect of repetitive transcranial magnetic stimulation(rTMS)on the recovery of neurological deficit in mice after ischemic brain injury by regulating the expression of DJ1.Methods A total of 30 healthy male C57BL/6J mice,aged 812 weeks,were randomly divided into shamoperation group(Sham group),ischemia/reperfusion group(MCAO group),and treatment group(rTMS group),with 10 mice in each group.The mice in the MCAO group were used to establish a mouse model of MCAO by the suture method,and those in the Sham group were given the same treatment as the MCAO group without suture occlusion.The mice in the Sham group and the rTMS group were treated with rTMS,and those in the MCAO group were not given any treatment.After 1 hour of modeling,the suture was extracted time,5 times a day for 7 consecutive days.Neurological deficit score was determined for each group of mice before and after treatment;TTC staining was used to measure infarct area;immunohistochemistry was used to measure the change in the expression level of DJ1 in neurons around the infarct area,and Western blotting was used to measure the change in the protein expression level of DJ1.Results Compared with the Sham group,the MCAO group and the rTMS group had a significant increase in neurological deficit score(2.80±0.63,2.10±0.32 vs 0.50±0.53,P<0.05),and compared with the MCAO group,the rTMS group had a significant reduction in neurological deficit score(P<0.05).Compared with the MCAO group,the rTMS group had a significant reduction in infarct area and a significant increase in the expression of DJ1(P<0.01).Conclusions In mice with ischemic brain injury,rTMS can effectively promote neurological function recovery and significantly reduce infarct area,possibly by up regulating the protein expression of neuronal DJ1 to intervene in the prognosis of such injury.
