摘要
目的研究紫斑牡丹Paeonia rocki i(S.G.Haw&Lauener)T.Hong&J.J.Li花粉化学成分及其体外抗氧化活性。方法紫斑牡丹花粉70%乙醇提取物的正丁醇部位采用硅胶柱、反相硅胶C 18柱、ODS、聚酰胺、Sephadex LH-20进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构。采用DPPH法评价其抗氧化活性。结果从中分离得到15个化合物,分别鉴定为柠檬黄素(1)、木犀草素-7-O-β-D-吡喃葡萄糖苷(2)、野漆树苷(3)、芹菜素-7-O-β-D-吡喃葡萄糖苷(4)、柠檬黄素-3-β-D-葡萄糖苷(5)、limocitrin-3-O-y1-β-D-sophoroside(6)、柯伊利素-7-O-β-D-吡喃葡萄糖苷(7)、白芍苷R1(8)、苯甲酰芍药苷(9)、白藜芦醇(10)、苯甲酸(11)、3-甲氧基-4-羟基苯甲酸(12)、没食子酸甲酯(13)、胡萝卜苷(14)、1,2,3,4,6-五没食子酰葡萄糖(15)。化合物1~2、10、15的IC 50值均低于0.02 mg/L,化合物15的IC 50值小于抗环血酸,化合物2的IC 50值与抗环血酸相当。结论化合物2~4、7~10、12、15均为首次从该植物中分离得到。化合物1~7、10、12~13、15均表现出一定的体外抗氧化性。
AIM To study the chemical constituents from the pollen of Paeonia rocki i(S.G.Haw&Lauener)T.Hong&J.J.Li and their in vitro antioxidant activities.METHODS The n-butanol fraction of 70%ethanol extract from the pollen of P.rockii was isolated and purified by silica gel column,reverse-phase silica gel C 18 column,ODS,polyamide and Sephadex LH-20,then the structures of obtained compounds were identified by physicochemical properties and spectral data.The in vitro antioxidant activities were evaluated by DPPH method.RESULTS Fifteen compounds were isolated and identified as limocitrin(1),luteolin-7-O-β-D-glucopyranoside(2),rhoifolin(3),apigenin-7-O-β-D-neospheroside(4),limocitrin-3-β-D-glucoside(5),limocitrin-3-O-y1-β-D-sophoroside(6),chrysoeriol-7-O-β-D-glucopyranoside(7),albiflorin R1(8),benzoylpaeoniflorin(9),resveratrol(10),benzoic acid(11),3-methoxy-4-hydroxybenzoic acid(12),methyl gallate(13),β-daucosterin(14),1,2,3,4,6-pentagalloylglucose(15).The IC 50 values of compounds 1-2,10,15 were lower than 0.02 mg/L.The IC 50 value of compound 15 was less than that vitamin C.The IC 50 value of compound 2 was the same with vitamin C.CONCLUSION Compounds 2-4,7-10,12,15 are isolated from this plant for the first time.Compounds 1-7,10,12-13,15 show certain in vitro antioxidant activities.
