摘要
目的 探讨去甲斑蝥素(NCTD)对人肝癌SMMC-7721细胞侵袭迁移作用及对EphA2/EphrinA1和E-cadherin表达的影响。方法 常规培养SMMC-7721细胞,取对数生长期细胞分为对照组及NCTD高、中、低剂量组,划痕实验检测细胞迁移能力,Transwell小室实验检测细胞侵袭能力,qRT-PCR和Western Blot分别检测细胞EphA2/EphrinA1和E-cadherin mRNA及蛋白质的表达情况。结果 NCTD明显抑制细胞侵袭和迁移,差异具有统计学意义(P<0.05);NCTD可以使肝癌细胞中EphA2、EphrinA1 mRNA及蛋白的表达量降低,E-cadherin mRNA和蛋白表达量升高,差异具有统计学意义(P<0.05)。结论去甲斑蝥素对人肝癌SMMC7721细胞迁移和侵袭有一定的抑制作用,其机制可能与NCTD下调EphA2/EphrinA1基因及蛋白的表达,引起E-cadherin上调,抑制肿瘤细胞EMT相关。
Objective To investigate the effects of norcantharidin(NCTD)on cell invasion and migration,the function of NCTD on expression of EphA2/EphrinA1 and E-cadherin were evaluated in HCC.Methods The HCC cell SMMC-7721 were cultured and treated with NCTD,including high,medium and low-dose.The scratch and transwell assays were used to detect the cell migration and invasion ability in HCC.The qRT-PCR and western blotting assays were used to confirm the expression of EphA2,EphrinA1 and Ephrina1,respectively.Results NCTD could significantly inhibit cell invasion and migration.The mRNA and protein levels of EphA2 and Ephrina1 were dramatically down-regulated by NCTD,while E-cadherin expressions were increased in HCC cell.Conclusion The mechanism of norcantharidin inhibiting the migration and invasion of SMMC7721 cell may be attributed to the downregulation of EphA2/EphrinA1 expression and(increase the level of E-cadherin to result in the inhibition of EMT in HCC.
