摘要
目的:为了探究中华补血草醇提物(Limonium sinense(Girard)Kuntze enthanol extract,LSKE)保肝的物质基础及其保肝作用机制。方法:采用乙醇加热回流提取得到中华补血草提取物(LSKE),通过高效液相色谱结合标准品鉴定LSKE的主要成分是木犀草素、山奈酚、槲皮素、杨梅素、圣草酚、异鼠李素6种黄酮类成分;通过酒精诱导小鼠急性肝损伤模型考察不同剂量中华补血草醇提物(100 mg/kg、200 mg/kg和400 mg/kg)对小鼠血清ALT、AST、SOD、GSH-Px和MDA含量及活性的影响;对小鼠肝脏组织病理切片进行H&E染色分析;并采用qRT-PCR和Western blot技术对炎症因子IL-1β、IL-6、TNF-α以及肝药酶CYP2E1的mRNA表达水平进行分析。结果:研究表明LSKE能够抑制血清中ALT、AST的水平,降低MDA水平,同时提高抗氧化酶SOD和GSH-Px的活性,以及抑制炎症因子和肝药酶CYP2E1的mRNA表达。结论:LSKE可通过改善因酒精引起的炎症水平、氧化应激水平和脂质过氧化水平发挥对小鼠酒精性肝损伤的保护作用。
Objective:To explore the material basis and mechanism of the Limonium sinense(Girard)Kuntze enthanol extract(LSKE)on liver protection.Methods:The active ingredients(LSKE)was extracted by ethanol heating from Limonium sinense(Girard)Kuntze.The main components of LSKE were identified by high performance liquid chromatography(HPLC)combined with standard substances as luteolin,kaempferol,quercetin,myricetin,sage phenol and isorhamnetin;The effects of different doses of LSKE(100 mg/kg,200 mg/kg and 400 mg/kg)on the contents and activities of serum ALT,AST,SOD,GSH-Px and MDA in mice were investigated by alcohol induced acute liver injury model;The pathological sections of mouse liver were analyzed by H&E staining;The inflammatory factor IL-1β,IL-6,TNF-αand liver drug enzyme CYP2E1 were detected by qRT-PCR and Western blot technology.Results:The results showed that LSKE could inhibit the levels of ALT and AST in serum,reduce the level of MDA,increase the activities of antioxidant enzymes SOD and GSH-Px,and inhibit the mRNA expression of inflammatory factor and liver drug enzyme CYP2E1.Conclusion:LSKE could protect mice from alcoholic liver injury by improving the level of inflammation,oxidative stress and lipid peroxidation caused by alcohol.
