橘小实蝇、瓜实蝇和南亚实蝇卵共生菌多样性及差异分析

Diversity and difference of symbiotic bacteria isolated from eggs of Bactrocera dorsalis,Zeugodacus cucurbitae and Zeugodacus tau

摘要 为探究橘小实蝇、瓜实蝇和南亚实蝇卵共生菌群落结构、多样性及其在不同种群间的差异,利用Illumina NovaSeq 6000测序技术分析3种实蝇不同种群卵共生菌的16S rDNA V3-V4高变区序列。结果表明,7个种群的实蝇卵共生菌序列共注释3 495个操作分类单元(OTUs),注释到界、门、纲、目、科和属水平的比例分别为96.08%、76.37%、74.48%、70.99%、61.09%和41.46%。放线菌门细菌主要在南亚实蝇卵中聚集,拟杆菌门细菌主要在橘小实蝇卵中聚集,而厚壁菌门和变形菌门细菌在3种实蝇卵中均有聚集。从3种实蝇实验室种群和橘小实蝇福州种群6~10代卵共生菌中筛选到相对丰度存在差异的细菌属为沙雷氏菌属、普罗威登斯菌属、漫游球菌属、Cosenzaea、米勒氏菌属、乳球菌属和肠球菌属,其中,沙雷氏菌属、普罗威登斯菌属、肠球菌属和乳球菌属为3种实蝇卵共生菌的优势属。此外,橘小实蝇安顺种群与橘小实蝇福州种群第10代、南亚实蝇安顺种群、南亚实蝇实验室种群的卵共生菌Simpson指数和Shannon指数差异显著。本研究可为3种实蝇引诱活性菌筛选及其功能研究提供参考。 To investigate the community structure,diversity,intraspecific and interspecific differences of symbiotic bacteria isolated from eggs of 3 kinds of fruit flies(Bactrocera dorsalis Hendel,Zeugodacus cucurbitae Coquillett and Zeugodacus tau Walker)with different populations,the 16S r DNA V3-V4 hypervariable regions of their symbiotic bacteria in eggs were sequenced using Illumina Nova Seq 6000 sequencing technology.A total of 3495 operational taxonomic units(OTUs)were annotated from the symbiotic bacte-ria in eggs of 7 fruit fly populations,covering 96.08%kingdom,76.37%phylum,74.48%class,70.99%order,61.09%family and 41.46%genus,respectively.Actinobacteria mainly aggregated in the eggs of Z.tau while Bacteroidota concentrated at those of B.dorsalis,and Firmicutes and Proteobacteria were commonly distributed in the eggs of all 3 fruit flies.In the experimental populations of the 3 kinds of fruit flies and B.dorsalis Fuzhou populations F6-F10,the symbiotic bacteria with relative abundance differences be-longed to Serratia,Providencia,Vagococcus,Cosenzaea,Moellerella,Lactococcus and Enterococcus,among which Serratia,Provi-dencia,Enterococcus and Lactococcus were the dominant genera.There were significant differences in the Simpson index and Shannon index of symbiotic bacteria between B.dorsalis Anshun population and Fuzhou population F10 as well as the counterpart of Z.tau An-shun population and its laboratory population.This study provided a reference for the screening of attractant active bacteria and their functional research in fruit flies.
