摘要
目的探讨白虎加人参汤对高脂饮食诱导的骨骼肌特异性胰岛素样生长因子1受体功能缺失(loss of skeletal muscle-specific insulin-like growth factor-1 receptor function,MKR)小鼠白色脂肪组织(white adipose tissue,WAT)炎症及肝脏脂肪变性的影响。方法30只8周龄雄性MKR小鼠高脂饮食诱导12周,成功复制肥胖症合并2型糖尿病小鼠模型后,随机分为模型组、白虎加人参汤组、二甲双胍组;另设10只同龄雄性FVB小鼠为正常对照组,各组小鼠予以相应药物或蒸馏水连续灌胃6周。末次给药禁食不禁水12 h后,检测空腹血糖(fasting blood glucose,FBG)并进行口服糖耐量试验(oral glucose tolerance test,OGTT),然后麻醉取材。常规生化法检测肝功能、血脂水平;苏木精-伊红染色法观察肝脏组织及附睾白色脂肪组织形态结构及病理变化;油红O染色及Masson染色观察肝脏组织脂质沉积及纤维化改变;酶联免疫吸附测定法检测血清白细胞介素1β(interleukin-1β,IL-1β)、白细胞介素6(interleukin-6,IL-6)、肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)水平;蛋白免疫印迹法(Western Blot)检测附睾白色脂肪组织IL-1β、IL-6、TNF-α蛋白相对表达量。结果糖脂代谢相关指标结果显示,与正常对照组比较,模型组FBG水平、OGTT曲线下面积及总胆固醇(total cholesterol,TC)、甘油三酯(triglycerides,TG)、低密度脂蛋白胆固醇(low-density lipoprotein cholesterol,LDL-C)含量明显增高(P<0.01),高密度脂蛋白胆固醇(high-density lipoprotein cholesterol,HDL-C)含量明显降低(P<0.01),而白虎加人参汤能明显改善肥胖MKR糖尿病小鼠上述指标(P<0.01)。肝脏脂肪变性相关指标结果显示,与正常对照组比较,模型组出现明显脂质沉积、轻度炎症细胞浸润及肝细胞纤维化,且模型组谷丙转氨酶(alanine aminotransferase,ALT)、谷草转氨酶(aspartate transaminase,AST)水平均明显增高(P<0.01);与模型组比较,白虎加人参汤组肝脏�
Objective To investigate the effects of Baihu Jia Renshen Decoction(BJRD)on white adipose tissue(WAT)inflammation and hepatic steatosis in MKR(loss of skeletal muscle-specific insulin-like growth factor-1 receptor function)mice induced by high-fat diet.Methods Thirty 8-week-old male MKR mice were fed a high-fat diet for 12 weeks and the mice model of obesity combined with type 2 diabetes mellitus was successfully replicated,which were randomly divided into model group,BJRD group and Metformin group.Ten male FVB mice of the same age were set up as normal control group.Mice in each group were given corresponding drugs or distilled water(model group and control group)by gavage for 6 consecutive weeks.Mice were fasted after the last administration,but water was provided ad libitum.Twelve hours after the last treatment,fasting blood glucose(FBG)and oral glucose tolerance test(OGTT)were performed.Then the mice were anesthetized,and the samples were collected.The biochemical method was used to detect liver function and serum lipid levels.Hematoxylin-eosin staining was used to observe the morphological structure and pathological changes of liver tissue and WAT of epididymis.Oil red O staining and Masson staining were used to observe lipid deposition and fibrotic changes of liver tissue.Serum levels of interleukin-1β(IL-1β),interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)were detected by enzyme-linked immunosorbent assay.The relative expressions of IL-1β,IL-6 and TNF-αin WAT of epididymis was measured by Western Blot.Results The results of glycolipid metabolism-related indexes showed that,compared with the normal control group,the FBG level,area under the OGTT curve,total cholesterol(TC),triglycerides(TG),and low-density lipoprotein cholesterol(LDL-C)level were significantly increased in the model group(P<0.01).High-density lipoprotein cholesterol(HDL-C)level was significantly decreased(P<0.01).It was found that BJRD significantly improved the above-mentioned indexes in obese diabetic MKR mice(P<0.01).The resul
