摘要
为降低发酵成本,提高红平红球菌(Rhodococcus erythropolis)XH-1发酵产海藻糖脂的能力,采用单因素试验及Box-Behnken试验确定其最佳培养基组成。结果表明,最佳培养基配方为豆饼粉14.0 g/L,甘油36.2 g/L,酵母浸粉0.2 g/L,KH2PO42.0 g/L,K2HPO42.0 g/L,NaCl 1.0 g/L,KNO_(3)1.0 g/L;最优发酵条件为初始pH 9.0、发酵温度30℃、摇床转速150 r/min、装液量45 m L/250 m L、接种量3%、种龄60 h。在此条件下,菌株XH-1的海藻糖脂产量(8.34 g/L)是优化前的5.59倍。理化性质分析结果表明,海藻糖脂有较高的表面活性和乳化能力,很强的耐热(100℃、2 h)、耐酸碱(pH 2~10)及耐盐(0~20%NaCl)性。该研究结果为海藻糖脂的大规模工业化生产与应用提供理论支持。
In order to reduce the fermentation cost and improve the trehalolipid production by Rhodococcus erythropolis XH-1,the optimal medium composition was determined by single factor tests and Box-Behnken tests.Results showed that the optimal medium formula were as follows:soybean cake powder 14.0 g/L,glycerin 36.2 g/L,yeast extract powder 0.2 g/L,KH2PO42.0 g/L,K2HPO42.0 g/L,NaCl 1.0 g/L,and KNO_(3)1.0 g/L.The optimum fermentation conditions were as follows:initial pH 9.0,fermentation temperature 30℃,shaking speed 150 r/min,liquid volume 45 ml/250 ml,inoculum 3%,and seed age 60 h.Under these optimal conditions,the trehalolipid yield of strain XH-1(8.34 g/L)was 5.59 times that of before optimization.The results of physicochemical property analysis showed that the trehalolipid had higher surface activity and emulsifying ability,and had strong thermal stability(100℃,2 h),acid-base stability(pH 2-10),and salt tolerance(0-20%NaCl).The research results provided theoretical support for the large-scale industrial production and application of trehalolipid.
