摘要
目的探究化学消融肾交感神经(RDN)对腹主动脉缩窄心力衰竭(简称“心衰”)大鼠心肌细胞线粒体功能的影响。方法采用腹主动脉缩窄法(TAC)建立大鼠慢性心衰模型,将心衰大鼠分为心衰组(n=3,暴露双侧肾动脉但不消融肾交感神经),RDN组(n=3,RDN术8周后腹腔注射生理盐水),Nogo-B组(n=4,RDN术8周后腹腔注射Nogo-B),NEP组(n=3,RDN术8周后腹腔注射NEP-40)。对肾动脉行HE染色并检测各组大鼠心肌细胞ATP含量及Tom-20表达量。结果与心衰组比较,RDN后大鼠肾动脉外膜神经纤维明显减少。RDN后各注药组肾动脉周围神经纤维数目由多到少依次为:NEP组、RDN组、Nogo-B组;RDN、Nogo-B及NEP组大鼠ATP浓度及Tom-20相对表达量均较心衰组大鼠升高,差异有统计学意义(P<0.05),RDN、Nogo-B、NEP三组间ATP含量及Tom-20相对表达量差异无统计学意义(P>0.05)。结论RDN可以改善心衰大鼠心肌细胞线粒体功能。
Objective To investigate the effects of chemical renal denervation(RDN)on mitochondrial function of cardiomyocytes in rats with heart failure(HF)induced by ab dominal aortic constriction.Methods A rat model of chronic HF was established by transverse aortic constriction(TAC)method,and rats with HF were divided into a HF group(n=3,with exposure of bilateral renal arteries but without RDN),a RDN group(n=3,injected intraperitoneally with normal saline 8 weeks after RDN),a Nogo-B group(n=4,injected intraperitoneally with Nogo-B 8 weeks after RDN),and a neutral endopeptidase(NEP)group(n=3,injected intraperitoneally with NEP-40 after 8 weeks of RDN).The renal arteries of rats were subjected to hematoxylin-eosin(HE)staining for test of the adenosine triphosphate(ATP)content and Tom-20 expression in cardiomyocytes in each group.Results Compared with the heart failure group,RDN significantly reduced the number of nerve fibers in the renal artery adventitia of rats.The number of nerve fibers around the renal artery in each injection group after RDN was in descending order:NEP group,RDN group,and Nogo-B group;The ATP concentration and relative expression of Tom-20 were higher in the RDN,Nogo-B and NEP groups than in the HF group,with statistically significant differences(P<0.05).However,there were no statistically significant differences among RDN,Nogo-B and NEP groups in ATP content and relative expression of Tom-20(P>0.05).Conclusion RDN can improve the mitochondrial function of cardiomyocytes in rats wi th HF.
