垂体腺瘤环状RNA Hsa_circ_0043459表达水平与术后复发的相关性 被引量:1

摘要 目的探讨垂体腺瘤组织中环状RNA hsa_circ_0043459的表达与术后复发的关系。方法回顾性分析2014年6月至2019年6月经手术治疗的120例垂体腺瘤患者的临床资料,另外选取20例同期经颅脑损伤内减压术中切除的正常脑组织。实时荧光定量PCR法检测垂体腺瘤及脑组织中hsa_circ_0043459的表达。垂体腺瘤患者均随访至2021年6月,按随访结局分为复发组和未复发组。多因素回归分析判定术后复发相关因素。结果垂体腺瘤组织中hsa_circ_0043459表达高于正常脑组织(P<0.05);术后2年内复发23例,复发率19.17%。相较于未复发组,复发组hsa_circ_0043459表达明显升高(P<0.05)。以120例垂体腺瘤hsa_circ_0043459的平均表达水平(3.26)为标准,分为高表达49例,低表达71例。高表达组复发率(36.73%)明显高于低表达组(7.04%)(P<0.05)。Kaplan-Meier生存曲线分析显示,高表达组术后复发时间明显较低表达组缩短(P<0.05)。多因素Logistic回归分析显示,hsa_circ_0043459表达升高,被认为是垂体腺瘤术后复发的独立危险因素(P<0.05)。结论hsa_circ_0043459在垂体腺瘤组织中表达升高,且与垂体腺瘤术后复发呈正相关。 Objective To explore the correlation between the expression level of the circular RNA(circRNA)hsa_circ_0043459 in pituitary adenoma(PA)and postoperative recurrence.Methods From June 2014 to June 2019,the clinical data of 120 surgically treated PA patients were retrospectively analyzed.Their PA tissues were collected for analysis(PA group).During the same period,20 normal brain tissues surgically collected from the decompression surgery for traumatic brain injury were collected as controls(control group).The expression levels of hsa_circ_0043459 in PA and control group were detected by real-time fluorescence quantitative polymerase chain reaction(qRT-PCR).PA patients were followed up until June 2021,and they were assigned into the recurrence group and the non-recurrence group according to follow-up data.Multivariate regression analysis was performed to determine the risk factors for the postoperative recurrence of PA.Results Hsa_circ_0043459 in PA group was significantly higher than that in control group(P<0.05),which in the recurrence group was significantly higher than that in the non-recurrence group(P<0.05).Within 2 years after surgery,there were 23(19.17%)cases of recurrence.According to the mean expression level of hsa_circ_0043459(3.26)in 120 PA patients,they were divided into high expression group(n=49)and low expression group(n=71).The recurrence rate in the high expression group was significantly higher than that in the low expression group(36.73%vs 7.04%,P<0.05).Kaplan-Meier survival curve showed that the postoperative recurrence time in the high expression group was significantly shorter than that in the low expression group(P<0.05).Multivariate logistic regression analysis showed that the upregulation of hsa_circ_0043459 was an independent risk factor for postoperative recurrence of PA(P<0.05).Conclusion Hsa_circ_0043459 is upregulated in PA patients,which is positively correlated with postoperative recurrence.
