摘要
目的探讨转铁蛋白受体(TFRC)对宫颈癌细胞增殖、迁移、侵袭、上皮-间充质转化的影响及可能的作用机制。方法慢病毒构建敲低和过表达TFRC的宫颈癌细胞系,并分为空白对照组、TFRC敲低阴性对照组、TFRC敲低组、TFRC过表达阴性对照组及TFRC过表达组。用蛋白印迹分析法检测TFRC、真核起始因子(EIF4A3)、E-钙黏蛋白(E-cadberin)、N-钙黏蛋白(N-cadherin)、波形蛋白(Vimentin)和锌指转录因子(Snail)的表达情况,通过平板克隆、细胞划痕和Transwell侵袭实验检测宫颈癌细胞的增殖、迁移和侵袭能力。结果与空白对照组相比,TFRC敲低组EIF4A3、N-cadherin、Vimentin和Snail蛋白表达水平下降、E-cadberin蛋白表达水平升高,细胞克隆数减少、迁移率下降和侵袭细胞数量减少(P均<0.05);相反,TFRC过表达组EIF4A3、N-cadherin、Vimentin和Snail表达量增加、E-cadberin表达量减少,细胞克隆数增加、迁移迁移率升高和侵袭细胞数量增加(P均<0.05)。结论TFRC在宫颈癌中呈高表达,通过调节EIF4A3可促进宫颈癌的增殖、迁移、侵袭、上皮-间充质转化;过表达和敲低EIF4A3可相应抵消敲低和过表达TFRC对宫颈癌细胞恶性生物学行为产生的影响。
Objective To investigate the effects of transferrin receptor(TFRC)on the proliferation,migration,in⁃vasion and epithelial-mesenchymal transition(EMT)of cervical cancer cells and the possible mechanism of action.Meth⁃ods Cervical cancer cell lines with knock-down and overexpression of TFRC were constructed by lentivirus,and were di⁃vided into the blank control group,TFRC knock-down negative control group,TFRC knock-down group,TFRC overexpres⁃sion negative control group,and TFRC overexpression group.The expression of TFRC,EIF4A3,E-cadberin,N-cad⁃herin,Vimentin and Snail was detected by Western blotting.The proliferation,migration and invasion of cervical cancer cells were detected by plate cloning,cell Scratch healing and Transwell invasion assay.Results Compared with the blank control group,the protein expression levels of EIF4A3,N-cadherin,Vimentin,and Snail in cervical cancer cells in TFRC knock-down group decreased,the protein expression levels of E-cadberin increased,and the number of cell clones,migration and invasion cells decreased(all P<0.05).On the contrary,the expression levels of EIF4A3,N-cad⁃herin,Vimentin and Snail increased,the expression levels of E-cadberin decreased,the number of cell clones in⁃creased,migration rate increased,and the number of invasive cells increased in the TFRC overexpression group(all P<0.05).Conclusions TFRC is highly expressed in cervical cancer,and can promote the proliferation,migration,inva⁃sion and EMT of cervical cancer by regulating EIF4A3.Overexpression and knock-down of EIF4A3 can counteract the ef⁃fects of knock-down and overexpression of TFRC on the malignant biological behavior of cervical cancer cells.
