摘要
目的:利用转录组学测序筛选2型糖尿病模型大鼠差异表达基因,初步探讨其与气阴两虚型证候表现的关联性。方法:随机选取雄性SD大鼠18只作为空白组,其余大鼠采用高糖高脂饲料喂养和腹腔注射链脲佐菌素(STZ)方法制备2型糖尿病大鼠模型。于第6周根据血糖和体质量均衡原则将造模成功的31只大鼠分为模型组14只和药物佐证组17只,模型组大鼠继续高糖高脂饲料喂养,药物佐证组大鼠灌胃消渴灵2.2 g·kg^(-1)·d^(-1),实验持续至第21周。模型制备期间定期检测大鼠体质量、进食量、饮水量、精神状态评分、运动评分、足温、抓力、脉搏幅度、呼吸频率和舌象积分,评价大鼠生理体征,血糖仪检测各组大鼠空腹血糖(FBG)水平。末次给药后,麻醉大鼠,采血,分离血清,氧化酶法检测各组大鼠血清中甘油三酯(TG)和总胆固醇(TC)水平,直接法检测各组大鼠血清中低密度脂蛋白胆固醇(LDL-c)和高密度脂蛋白胆固醇(HDL-c)和水平,酶联免疫吸附测定(ELISA)法检测各组大鼠血清中CD4、CD8、环磷酸腺苷(cAMP)和环磷酸鸟苷(cGMP)水平。取大鼠肝组织,采用转录组学测序,筛选模型组与空白组和模型组与药物佐证组的差异表达基因。结果:与空白组比较,模型组大鼠生理体征基本符合气阴两虚的证候表现;与模型组比较,药物佐证组大鼠部分气阴两虚的生理体征明显改善。与空白组比较,模型组大鼠FBG水平和血清中TC、TG及LDL-c水平明显升高(P<0.01),HDL-c水平差异无统计学意义(P>0.05);与模型组比较,药物佐证组大鼠上述指标差异无统计学意义(P>0.05)。ELISA法检测,与空白组比较,模型组大鼠血清中CD4水平及CD4/CD8比值明显降低(P<0.01),CD8水平差异无统计学意义(P>0.05),cAMP水平及cAMP/cGMP比值明显升高(P<0.01),cGMP水平明显降低(P<0.01);与模型组比较,药物佐证组大鼠血清中CD4水平和CD4/CD8比值明显升高(P<0.01),cGMP水平
Objective:To screen the differentially expressed genes in type 2 diabetes mellitus model rats by transcriptomics sequencing,and to preliminarily explore the relationship between the differentially expressed genes and the syndrome manifestations of deficiency of both Qi and Yin.Methods:Eighteen male SD rats were randomly selected as control group,and the other rats were fed with high-fat and highsugar diet and injected with streptozotocin(STZ)in the abdomen to establish the type 2 diabetes models.On the 6th week,31 successfully modeling rats were divided into model group(n=14)and drug evidence group(n=17)according to the principle of blood glucose and body mass balance.The rats in model group were fed with high sugar and high fat diet sequentially.The rats in drug evidence group were given Xiaokeling 2.2 g·kg^(-1)·d^(-1) by gavage;the experiment last for 21 weeks.The body masses,food intakes,water intakes,mental state scores,sport scores,foot temperatures,grasping force,pulse amplitudes,respiratory rates,and tongue image integral of the rats were detected and the physiological signs of the rats during model preparation were evaluated.The fasting blood glucose(FBG)levels of the rats in various groups were detected by blood glucose meter.After the last administration,the rats were anesthetized,the blood samples were collected and the serum samples were separated.The levels of triglyceride(TG)and total cholesterol(TC)of the rats were detected by oxidase method.The levels of high density lipoprotein cholesterol(HDL-c)and low density lipoprotein cholesterol(LDL-c)of the rats were detected by direct method,and the levels of CD4,CD8,cyclic adenosine monophosphate(cAMP),and cyclic guanosine monophosphate(cGMP)of the rats were detected by enzyme-linked immunosorbent assay(ELISA)method.The liver tissue of the rats was obtained,and the differentially expressed genes between model group and control group and between model group and drug evidence group were screened by transcriptomic sequencing.Results:Compared with blank gr
