黑麦GIF基因家族全基因组鉴定及其表达分析 被引量:1

摘要 生长调节互作因子(GRF-interacting factor,GIF)是植物体内一类转录共激活因子,在植物生长发育过程中起重要作用。为挖掘和利用黑麦GIF家族基因,利用同源比对方法对黑麦GIF基因家族进行了鉴定,并系统分析了黑麦与其他禾本科植物及模式植物拟南芥GIF基因的系统发育关系、蛋白结构域、基因结构和共线性关系。结果显示,在黑麦基因组中共鉴定到3个GIF基因家族成员,分布在4R和5R染色体上;系统进化分析发现,黑麦GIF基因家族成员与小麦和大麦的亲缘关系更近。蛋白结构域和基因结构分析发现,GIF基因家族成员在黑麦及其他禾本科植物中的蛋白结构域和基因结构相对保守,均含有4个外显子。共线性分析发现,黑麦ScGIF1基因与其他禾本科植物GIF基因间均存在共线性关系,其中与小麦间共线基因数最多。Ka/Ks分析发现,黑麦GIF基因家族成员在进化中受到纯化选择。此外,在黑麦穗部鉴定到一个特异性高表达的GIF基因(ScGIF1),其编码的蛋白含有多个磷酸化位点,推测ScGIF1蛋白可能是通过磷酸化来调控黑麦麦穗的发育和粒型的。 GRF-interacting factor(GIF)gene family is a class of transcriptional co-activators in plants and plays an important role in plant growth and development.In order to explore and utilize GIF genes in rye,we systematically identified the rye GIF family at genome level in this study.Phylogenetic relationship,conserved domain,gene structure and collinearity of rye GIF gene were analyzed.The results showed that three members of GIF gene family were found in the rye genome,which were distributed on 4R and 5R chromosomes.Phylogenetic analysis showed that the three rye GIF genes are closer to GIF gene of wheat and barley.The protein domain and gene structure of GIF gene family in rye and other gramineous plants are relatively conservative,and they all contain four exons.It was found that there were collinear pairs between ScGIF1 genes and those in other gramineous plant genomes,and rye displayed the most pairs with wheat.Ka/Ks analysis showed that rye GIF genes family were purely selected during evolution.In addition,a GIF gene(ScGIF1)displayed the specifically high expression in the rye spikelet,and its protein contained multiple phosphorylation sites.It was speculated that the ScGIF1 protein may have functions in regulating spikelet development and affecting grain shape through phosphorylation in rye.
