摘要
目的建立同时分析血液中11种传统毒品及代谢物的液相色谱-串联质谱(LC-MS/MS)快速测定方法。方法血液经乙腈沉淀蛋白后用Phenomenex Kinetex Biphenyl 100Å(100 mm×3.0 mm×2.6µm)色谱柱分离,以11种毒品对应的氘代标品及甲氧那明作为内标,采用多反应监测模式同时测定血液中11种传统毒品及代谢物。结果各化合物检出限为0.5~2.0 ng/mL,定量限为1~5 ng/mL,在相应浓度范围内,11种传统毒品及代谢物均具有良好的线性关系,相关系数大于0.995,方法准确度为92.25%~109.56%,提取回收率为54.21%~101.86%,基质效应为79.60%~111.12%。结论该方法选择性强、灵敏度高、所需样品量少(100μL)、前处理过程简单快速(小于5 min),可用于毒驾等司法鉴定案件中11种传统毒品及代谢物的检验。
Objective To establish a rapid method for the simultaneous analysis of 11 traditional drugs and metabolites in blood by liquid chromatography mass spectrometry(LC-MS/MS).Methods The blood was precipitated with acetonitrile and then separated with Phenomenex Kinetex Biphenyl 100Å(100 mm×3.0 mm×2.6µm)chromatographic column,with internal standard of methoxyphenamine and deuterated standard corresponding to 11 drugs.Eleven traditional drugs and metabolites in blood was detected by multi reaction monitoring mode simultaneously.Results The limit of detection of each compound was 0.5-2.0 ng/mL and the limit of quantitative was 1-5 ng/mL.Within the corresponding concentration range,11 traditional drugs and metabolites had good linear relationship,the correlation coefficient was greater than 0.995,the accuracy of the method was in the range of 92.25%-109.56%,the recoveries were in the range of 54.21%-101.86%,and the matrix effect was 79.60%-111.12%.Conclusion The method has strong selectivity,high sensitivity and characterized by a low required sample volume(100μL),a simple and fast pretreatment process(less than 5 min).It can be used to detect 11 traditional drugs and metabolites in forensic cases such as drug driving.
