卵丘颗粒细胞中卵母细胞分泌因子表达水平对卵母细胞成熟度的影响 被引量:1

Expression levels of oocyte secreted factors in the cumulus cells and its relationship with oocyte maturation

摘要 目的评估单卵丘颗粒细胞(CCs)中GDF9、BMP15和TGF-β1的mRNA表达水平对卵母细胞成熟度的影响及在不同促排卵方案中的表达水平差异。方法收集直接行ICSI治疗的患者64例679份CCs,其中拮抗剂方案组CCs 272份,微刺激方案组CCs 245份,PPOS方案组CCs 162份,卵母细胞与单卵丘颗粒细胞一一对应。提取的mRNA表达水平采用real-time PCR方法进行检测,分析卵母细胞发育不同阶段卵母细胞分泌因子的含量,并比较不同促排卵方案中GDF-9、BMP-15和TGF-β1表达水平的差异。结果卵母细胞分泌因子GDF-9、BMP-15和TGF-β1的表达水平,MⅡ组均显著高于MⅠ组,MⅠ组显著高于GV组,正常受精组显著高于异常受精组,优质胚胎组显著高于非优质胚胎组。微刺激方案的Gn量较拮抗剂方案及PPOS方案更低有明显统计学差异。GDF-9、BMP-15和TGF-β1在三种促排卵方案中的表达量无明显统计学差异。结论GDF-9、BMP-15和TGF-β1可作为评价卵母细胞成熟度及其发育潜能的客观指标,其表达量与卵母细胞成熟度相关。在这三种促排卵方案中,微刺激方案的Gn量最低。CCs中的卵母细胞分泌因子的表达水平在这三种促排卵方案无明显差异,对卵母细胞成熟度的治疗效果相似。 Objective To evaluate the effect of the mRNA expression levels of oocyte-secreted factors(OSFs)on the oocyte maturity,expression levels of GDF-9,BMP-15,and TGF-β1 in individual cumulus cells(CCs)were analyzed,and compare differences in the expression levels in different protocols.Methods Collect 679 CCs from 64 patients directly undergoing tracytoplasmic sperm injection(ICSI),including 272 CCs in antagonist group,245 CCs in micro-stimulation group,162 CCs in progestin-primed ovarian stimulation(PPOS).Single-cumulus granulosa cells correspond one-to-one to the oocytes.The mRNA from single cumulus granulosa cells was extracted and assessed by real-time quantitative PCR,to analyze the expression quantity of oocyte-secreted factor in oocytes at different developmental stages,and compare the differences in the expression levels of GDF-9,BMP-15 and TGF-β1 in different ovulation induction protocol.Results The expression levels of oocyte secreted factors GDF-9,BMP-15 and TGF-β1 were significantly higher in MII oocytes than the M I oocytes,the MI group was significantly higher than the GV group,the normal fertilization group was higher than the abnormal fertilization group and the good-quality embryo group was significantly higher than the poor-quality embryo group.The total amount of Gn in micro-stimulation group was lower than the antagonist group and the PPOS group,and there are significant statistical differences.The expression levels of BMP-15,GDF-9 and TGF-β1 were not significant statistical differences in different protocols.Conclusion GDF-9,BMP-15 and TGF-β1 may be used as potential indicators for evaluating the oocytes maturity and the developmental potential,and are correlated with the oocyte maturity.The total amount of Gn in micro-stimulation group was the lowest in the three group.The gene expression level of oocyte-secreted factors in CCs may not different among the three groups,and have similar therapeutic effects for oocyte maturity.
