摘要
目的探究外源激素对浙贝母组织培养的影响并进行组织形态学观察。方法以浙贝母鳞茎为外植体,接种于附加不同外源激素组合的MS固体培养基上,研究外源激素对浙贝母愈伤组织诱导、增殖及不定芽分化的影响。结果浙贝母鳞茎作为外植体,适合愈伤组织诱导培养基为MS+KT 1.0 mg/L+NAA 1.0 mg/L,诱导率为64.2%;在增殖培养基MS+6-BA 1.0 mg/L+TDZ 0.15 mg/L中,愈伤组织增殖效果最好;而最佳不定芽分化培养基为MS+6-BA 1.0 mg/L+TDZ 0.15 mg/L,发生率为85.6%;在增殖培养基MS+6-BA 1.0 mg/L+IBA 0.1 mg/L中,不定芽的增殖效果最好。结论确定了浙贝母愈伤组织诱导、增殖及分化的培养条件,建立了浙贝母愈伤组织及不定芽的诱导增殖体系,为进一步开展脱毒苗培育和新品种选育奠定基础。
Objective To explore the effect of exogenous hormones on tissue culture of Fritillaria thunbergii and observe its histomorphology.Methods The bulbs of F.thunbergii were inoculated on MS solid medium with different combinations of hormones to study the effects of exogenous hormones on callus induction,proliferation and differentiation of adventitious buds.Results When the bulbs of F.thunbergii were used as explants,the suitable medium for callus induction was MS medium adding 1.0 mg/L KT and 0.1 mg/L NAA,with the induction rate of 64.2%.The optimum medium for callus proliferation was MS medium adding 1.0 mg/L 6-BA and 0.15 mg/L TDZ,while the suitable medium for callus redifferentiation was also the same,with the induction rate of 85.6%.The MS medium adding 1.0 mg/L 6-BA and 0.1 mg/L IBA was beneficial to the growth of adventitious buds.Conclusion This experiment established a way to induce callus of F.thunbergii,then differentiate into adventitious buds and proliferate.In the experiment,the culture conditions of callus proliferation and differentiation of F.thunbergii are determined.The induction and proliferation system of callus and adventitious buds of F.thunbergii is basically established,which laid a foundation for further development of virus-free seedling cultivation and new variety breeding.
