摘要
为了获得在农业生物防治具有广谱抗菌作用的芽孢杆菌类益生菌,经多轮分离筛选,确定了一株对多种致病菌均具有较好抑菌效果的BA-300菌株。经16S rDNA测序比对、形态特征观察和生理生化分析,鉴定菌株BA-300为贝莱斯芽孢杆菌(Bacillus velezensis)。为提高抑菌活性,对菌株BA-300发酵条件进行优化。从6种常见的细菌培养基中筛选出最佳培养基,对时间、温度和pH三种发酵条件进行优化。将稀释5倍和10倍的菌株BA-300发酵液分别喷洒到樱桃树上,结果表明,稀释5倍的发酵液实验组对樱桃树叶片细菌性穿孔病的防治效果可以达到78.4%,稀释10倍的实验组也能达到70.0%。与对照组相比,樱桃产量有明显提高,说明贝莱斯芽孢杆菌BA-300有较高的生物防治作用。
To obtain probiotics with broad-spectrum antibacterial effects in agricultural biological control,a BA-300 strain with good antibacterial effects against various pathogenic bacteria was identified.After 16S rDNA sequencing comparison,morphological observation,physiological and biochemical analysis,the strain BA-300 was identified as Bacillus velezensis.To improve antibacterial activity,the fermentation conditions of strain BA-300 were optimized.The optimal culture medium was selected from 6 common bacterial cultures and the fermentation conditions of time,temperature and pH were optimized.The fermentation broth of strain BA-300 was diluted 5 times and 10 times and sprayed on cherry trees,respectively.The results showed that the fermentation broth had a control effect of 78.4%in the group diluted by 5 times on bacterial perforation disease in cherry leaves,while the group diluted by 10 times could achieve 70.0%.Compared with the control group,the yield of cherry in the experimental group significantly increased,indicating that Bacillus subtilis BA-300 has a high biocontrol effect.
