摘要
为了优化抗感颗粒处方水提取工艺,并研究其作为饲料添加剂对养殖鲫鱼生长性能及免疫力的影响,试验在赤芍和绵马贯众提取的单因素试验基础上,选取3个考察因素,以芍药苷的质量浓度为响应值,利用Design-Expert V8.0.6软件进行因素和水平设计,采用Box-Behnken法设计响应面试验方案,将得出的结果进行模型拟合及方差分析,确定最优工艺后,进行3次重复验证试验。利用响应面试验方法优化的水提取工艺进行抗感颗粒处方水提物的提取,并在基础日粮中添加质量分数为2%的抗感颗粒处方水提物,制成膨化鱼饲料。将60尾鲫鱼分为正常组、变温对照组和变温试验组,每组20尾,正常组与变温对照组投喂普通膨化鱼饲料,变温试验组投喂等量的添加质量分数为2%的抗感颗粒处方水提物的膨化鱼饲料,进行60 d的养殖试验。试验结束后对试验鱼称重,统计增重率、特定生长率、肥满率及饲料效率等;采集血液,分离血清,分别检测血清球蛋白、血清总蛋白、丙氨酸氨基转移酶(ALT)、碱性磷酸酶(AKP)、天冬氨酸氨基转移酶(AST)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)及丙二醛(MDA)等指标;解剖试验鱼取心脏、肝脏、肾脏等并称重,计算脏器系数。结果表明:在单因素试验结果的基础上,选取提取时间、加水量、提取次数为考察因素,浸泡时间为2 h,响应面试验方法优化结果为加水量6倍,浸泡2 h,提取2次,每次1.5 h。采用此条件进行3次重复性试验验证,提取得到的芍药苷质量浓度分别为2.9816,2.5830,2.9695 mg/mL,平均值为2.8447 mg/mL,与模型预测值(2.2537 mg/mL)的差异较小。变温试验组的增重率、特定生长率、饲料效率及肝脏系数均显著高于变温对照组(P<0.05),且与正常组差异不显著(P>0.05)。变温试验组血清总蛋白质量浓度极显著高于变温对照组(P<0.01),血清球蛋白质量浓度显著高于变温对照组(P<0.05),�
In order to optimize the optimal scheme of water extraction process of Kanggan granule prescription,and to study its effect on growth performance and immunity of cultured crucian carp using as a feed additive,on the basis of the single-factor test of the extracted red peony root and Dryopteris Crassirhizoma,three investigation factors were selected;the mass concentration of peony glycoside was used as the response value,and the factor and level design were carried out by Design-Expert V8.0.6 software for the response surface test.The Box-Behnken method was used to design the response surface test protocol,and the results obtained were performed for model fitting and variance analysis.After the optimal process was determinined,3 replicate validation tests were performed.The response surface test method was used to optimize the water extraction process for the extraction of water extracts of Kanggan granule prescription;water extracts of Kanggan granule prescription was added to the basal diet with a mass fraction of 2%to make puffed fish feed.Sixty crucian carp were divided into normal group,variable-temperature control group and variable-temperature test group with 20 fish in each group.The normal group and the variable-temperature control group were fed with normal puffed fish feed,and the variable-temperature test group was fed with equal amount of puffed fish feed supplemented with 2%mass fraction of water extracts of Kanggan granule prescription for the breeding test of 60 d.At the end of the experiment,the fish were weighed,and the weight gain rate,specific growth rate,fattening rate and feed efficiency were counted;the blood was collected,serum was separated,and serum globulin,serum total protein,alanine aminotransferase(ALT),alkaline phosphatase(AKP),aspartate aminotransferase(AST),superoxide dismutase(SOD)and catalase(CAT),malondialdehyde(MDA),etc.were measured.The heart,liver and kidney of the fish were taken and weighed in the anatomical test to calculate the organ coefficients.The results showed that b
