摘要
目的探讨瘦素对人乳腺癌MCF-7细胞增殖与迁移的影响。方法瘦素处理MCF-7细胞,MTT与划痕实验检测瘦素对MCF-7细胞增殖与迁移的影响,Western blotting和qPCR分别检测瘦素对MCF-7细胞MMP14蛋白及基因表达的影响。结果MTT检测结果显示,不同浓度瘦素处理组MCF-7细胞增殖均发生明显增殖,且具有浓度依赖性;划痕实验结果显示,瘦素处理乳腺癌细胞的迁移能力明显升高;Western blotting和qPCR检测结果显示,瘦素可上调MCF-7细胞MMP14蛋白与基因表达水平,并随着瘦素浓度的增加而升高,且100 ng/ml以上瘦素浓度处理差异显著(P<0.05)。结论瘦素通过上调人乳腺癌MCF-7细胞MMP14的表达,从而促进乳腺癌细胞的增殖与迁移。
Objective To investigate the effect of leptin on the proliferation and migration of human breast cancer MCF-7 cells.Methods MCF-7 cells were treated with leptin.The effect of leptin on the proliferation and migration of MCF-7 cells was detected by MTT and scratch assay.The effect of leptin on the expression of MMP14 protein and gene in MCF-7 cells was detected by Western blotting and qPCR.Results The MTT assay showed that different concentrations of leptin could promote the proliferation of MCF-7 cells in a concentration dependent manner.Scratch test results showed that the migration ability of breast cancer MCF-7 cells increased significantly in the leptin treated group.Western blotting and qPCR showed that leptin could up-regulate the expression of MMP14 protein and gene in MCF-7 cells,and it increased depending on leptin concentration,and the concentration of leptin above 100 ng/ml could cause significant difference(P<0.05).Conclusion Leptin could promote the proliferation and migration of breast cancer cells by up-regulating the expression of MMP14 in MCF-7 cells.
