摘要
目的探讨轴蛋白(AXIN)与β-连环蛋白(β-catenin)在卵巢子宫内膜异位症(OE)恶变过程中的表达及临床意义。方法采用定量酶联免疫吸附试验(ELISA)及免疫组化方法,检测30例OE组织、17例非典型子宫内膜异位症(aEM)组织及30例子宫内膜异位症相关性卵巢癌(EAOC)组织中AXIN与β-catenin的表达情况,并分析β-catenin及AXIN与EAOC病人的年龄、FIGO分期、病理组织类型、肿瘤侧别等临床特征的关系。结果免疫组化检测显示,AXIN在OE组、aEM组、EAOC组的阳性表达率分别为76.7%(23/30)、41.2%(7/17)、30.0%(9/30),差异有统计学意义(χ^(2)=13.852,P<0.05);β-catenin在OE组、aEM组、EAOC组的阳性表达率分别为13.3%(4/30)、29.4%(5/17)、63.3%(19/30),差异有统计学意义(χ^(2)=16.661,P<0.05)。ELISA检测显示,AXIN在OE组、aEM组、EAOC组的表达水平逐渐降低,差异有统计学意义(F=1986.939,P<0.001);β-catenin在OE组、aEM组、EAOC组的表达水平逐渐升高,差异有统计学意义(F=7361.889,P<0.001)。EAOC病变组织中AXIN与β-catenin的表达呈负相关(r=-0.86,P<0.05);AXIN及β-catenin的表达与肿瘤FIGO分期、组织学分级有显著相关性(P=0.000~0.013)。结论AXIN低表达与β-catenin高表达可能参与了OE的恶变过程,且可能促进了EAOC的浸润转移。
Objective To investigate the expression and clinical significance of AXIN andβ-catenin in the malignant transformation of ovarian endometriosis(OE).Methods ELISA and immunohistochemistry were used to measure the expression of AXIN andβ-catenin in 30 samples of OE,17 samples of atypical endometriosis(aEM),and 30 samples of endometriosis-associated ovarian cancer(EAOC),and the association ofβ-catenin and AXIN with the clinical features of EAOC was analyzed,including age,FIGO stage,pathological type,and tumor side.Results Immunohistochemistry showed that the positive expression rate of AXIN was 76.7%(23/30)in the OE group,41.2%(7/17)in the aEM group,and 30.0%(9/30)in the EAOC group,and there was a significant difference between the three groups(χ^(2)=13.852,P<0.05);the positive expression rate ofβ-catenin was 13.3%(4/30)in the OE group,29.4%(5/17)in the aEM group,and 63.3%(19/30)in the EAOC group,and there was also a significant difference between the three groups(χ^(2)=16.661,P<0.05).ELSIA showed that the expression level of AXIN gradually decreased in the OE group,the aEM group,and the EAOC group(F=1986.939,P<0.001),while the expression level ofβ-catenin gradually increased in the OE group,the aEM group,and the EAOC group(F=7361.889,P<0.001).The expression of AXIN was negatively correlated with that ofβ-catenin in EAOC lesion(r=-0.86,P<0.05),and the expression of AXIN andβ-catenin was significantly correlated with FIGO stage and histological grade(P=0.000-0.013).Conclusion The low expression of AXIN and the high expression ofβ-catenin may be involved in the malignant transformation of OE and might even promote the invasion and metastasis of EAOC.
