摘要
目的基于生物信息学分析miR-143调控Toll样受体2(TLR2)信号通路对溃疡性结肠炎(UC)的影响。方法在GEO数据库中下载GSE75214、GSE53306、GSE48958基因数据集及miRNA数据集GSE68306进行分析。将符合标准的差异miRNA用miRDB软件预测靶基因并与筛选的差异基因映射,挑选出UC中关键miRNA进行实验。按随机数字表法将20只小鼠分为正常组及UC组,每组10只。用葡聚糖硫酸钠进行造模,苏木精-伊红染色法观察病理变化,ELISA检测肿瘤坏死因子-α(TNF-α)含量,Western blot检测结肠TLR2、白细胞介素-1β(IL-1β)蛋白含量,实时荧光定量PCR检测结肠组织miR-143、核转录因子-κB(NF-κB)、髓样分化因子(MyD88)含量。结果通过分析发现,共85个差异基因(包括34个下调及51个上调)及19个差异miRNA(5个DEMs高表达,14个DEMs低表达),其中miR-143为关键miRNA。实验显示,与正常组比较,UC组血清中TNF-α含量显著升高,结肠组织miR-143表达显著降低(P<0.05),TLR2、IL-1β蛋白表达明显升高,NF-κB、MyD88 mRNA含量升高(P<0.05)。结论miR-143为UC的重要miRNA,其可能通过调控TLR2通路影响UC发生发展。
Objective To explore the effects of miR-143 regulation of Toll-like receptor 2(TLR2)signaling pathway on ulcerative colitis(UC)based on bioinformatics.Methods Gene data sets GSE75214,GSE53306,GSE48958 and miRNA data set GSE68306 were downloaded from GEO database for analysis.The target genes of the differential miRNAs meeting the criteria were predicted by miRDB software,and mapped with the screened differential genes,and the key miRNA in UC were selected for experiments.According to random number table method,20 mice were divided into normal group and UC group,with 10 mice in each group.Sodium gluconosulfate was used for modeling.The pathological changes were observed by hematoxylin-eosin staining staining.The contents of tumor necrosis factor-α(TNF-α)were detected by ELISA,and the contents of TLR2 and interleukin-1β(IL-1β)in colon were detected by Western blot.The contents of miR-143,nuclear transcription factor-κB(NF-κB),and myeloid differentiation factor(MyD88)in colon tissue were determined by real-time quantitative PCR.Results There were 85 DEMs,including 34 DEMs were downregulated and 51 DEMS were up-regulated,and 19 DEMS(5 DEMS were highly expressed,and 14 DEMS were underexpressed),with miR-143 being the key miRNA.The results showed that compared with normal group,the serum TNF-αcontent in UC group was significantly increased,the expression of miR-143 in colon tissue was significantly decreased(P<0.05),the protein expressions of TLR2 and IL-1βwere significantly increased,and the mRNA contents of NF-κB and MyD88 were significantly increased(P<0.05).Conclusion miR-143 is an important miRNA in ulcerative colitis,which may affect the development of UC through regulation of TLR2 pathway.
