吐曼河河水可培养功能菌株的筛选与生化初步鉴定

摘要 通过对吐曼河河水功能菌株的分离纯化,共得到30株放线菌,48株细菌,12株产淀粉酶菌。共106株菌株经革兰氏染色86株为阳性菌。在酶学特性研究中,得到产过氧化氢酶97株,产淀粉酶75株;78株菌株参与产蛋白酶的测定中得到产蛋白酶36株。在106株菌种50株菌能够充分利用脂酶Tween20为底物生长;59株能够利用Tween40为底物生长;78株菌株中共得到有48株利用Tween80生长。通过淀粉酶筛选培养基分离出5株菌能在淀粉培养基上产生透明圈较大的菌株,其中测量D3菌株的透明圈D/d值为3,D5为3.25 cm,D10为2.9 cm,D11为3.12 cm,D28为3.2 cm。 A total of 30 strains of actinomycetes,48 strains of bacteria and 12 strains of amylase-producing bacteria were obtained through the isolation and purification of functional strains from the river water of Tuman River.A total of 106 strains were positive by Gram staining 86 strains.In the study of enzymatic properties,97 strains of peroxidase-producing bacteria and 75 strains of amylase-producing bacteria were obtained;78 strains of bacteria were involved in the determination of protease-producing bacteria and 36 strains of protease-producing bacteria were obtained.In 106 strains 50 strains can make full use of lipase Tween20 as substrate growth;59 strains can use Tween40 as substrate growth;78 strains in total got 48 strains using Tween80 growth.Five strains were isolated by amylase screening medium to produce a larger clear circle on starch medium,where the clear circle D/d value of strain D3 was measured as 3 cm,D5 as 3.25 cm,D10 as 2.9 cm,D11 as 3.12 cm and D28 as 3.2 cm.The genomic DNA of the strains was extracted and the products after PCR amplification were observed under gel imager and 17 of them were found to have bright bands,indicating that 17 amplification products were successfully obtained.
