摘要
为了对新疆某牛场结核病进行筛查,并确定有病变的结核阳性牛以分离培养牛分枝杆菌,本试验采用比较皮内变态反应试验、牛结核菌素点眼试验、γ-干扰素试验和牛分枝杆菌抗体ELISA试验对新疆某牛场的1 185头牛进行检测,锁定疑似结核阳性牛并进行剖检,将结核疑似病料分离菌株并鉴定。结果显示:选取的3头4种检测方法皆为阳性的牛在淋巴结、胸壁或脾脏有明显病理学变化,综合PCR检测结果和全基因组测序结果,鉴定为牛分枝杆菌。结果表明,使用针对不同感染阶段的多种检测方法可以更准确地找到结核阳性病变牛,肺外结核仍普遍存在于新疆某牛场中。
In order to screen for tuberculosis(TB)in cattle farm in Xinjiang and identify the TB positive cattle with pathological changes for isolation and culture of Mycobacterium bovis(M.bovis),this study conducted comparative intradermal allergy test,bovine tuberculin eyeshadow test,γ-interferon test and M.bovis antibody ELISA test on 1185 cattle from a cattle farm in Xinjiang.The cattle suspected of tuberculosis were identified and samples were cultured for M.bovis isolation.The results showed that 3 cows that were positive for the 4 detection methods had significant pathological changes in lymph nodes,chest wall or spleen,and the resulting isolates were identified as M.bovis by PCR and whole genome sequencing.Thus,the detection methods for different stages of M.bovis infection allow accurate identification of TB positive cattle,and confirm the widespread presence of extrapulmonary tuberculosis in cattle farms in Xinjiang.
