摘要
通过网络药理学及实验探究广藿香抗氧化和美白作用机制。先在TCMSP数据库收集广藿香活性成分及其靶点,然后在String平台进行PPI分析并找出关键靶点,利用Cytoscape3.7.1构建“广藿香-活性成分-靶点”图,基于Metascape数据库进行KEGG和GO富集分析。通过实验,对比水提、75%乙醇提取下,提取物的活性和有效成分含量,确定广藿香的提取工艺。结果显示,收集到活性成分11余种,靶点169个;关键信号通路包括细胞衰老等。通过广藿香对自由基清除实验和抑制酪氨酸酶实验证实,广藿香具有抗氧化应激和美白等作用,75%醇提法为优势提取工艺。
To investigate the antioxidant and whitening mechanism of patchouli through network pharmacology and experiments.The active ingredients and targets of patchouli were collected in the TCMSP database,PPI analysis was performed on the String platform to identify the key targets,the“patchouli-active ingredient-target”map was constructed by Cytoscape 3.7.1,and KEGG and GO enrichment analysis were performed based on the Metascape database.Through experiments,the extraction process of patchouli was determined by comparing the active and active ingredient content of the extract under water extraction and 75%ethanol extraction.More than 11 active ingredients and 169 targets were collected.Key signaling pathways include cellular senescence.Through patchouli's free radical scavenging experiment and tyrosinase inhibition experiment,it was confirmed that patchouli has antioxidant stress and whitening effects,and 75%alcohol extraction is the dominant extraction process.
