运动结合生慧汤对睡眠剥夺大鼠海马神经元树突棘发育异常和认知损害的影响 被引量:1

Effects of Exercise Combined with Shenghui Decoction on Abnormal Development of Hippocampal Neuronal Dendritic Spines and Cognitive Impairment in Sleep Deprived Rats

摘要 目的 探讨运动结合生慧汤是否可以协同增效的防止睡眠剥夺大鼠学习记忆损伤,及其可能的作用机制。方法 将SD大鼠50只随机分为空白组(C组)、模型组(M组)、模型+运动组(ME组)、模型+生慧汤组(MS组)及模型+运动组+生慧汤组(MES组),ME组进行4周的跑台运动,MS组进行4周的生慧汤灌胃,而MES组同时进行4周的跑台运动和生慧汤灌胃之后,使用多平台水环境进行72 h的睡眠剥夺。睡眠剥夺结束后采用Morris水迷宫检测各组大鼠的学习记忆能力,利用HE染色和Golgi染色观察大鼠海马区神经元形态、树突分支棘单位树突棘密度的变化,使用免疫蛋白印记法检测海马CA3区PSD95、SYN1、GAP43及RAC1蛋白表达水平。结果 与C组比较,M组大鼠平台潜伏期和游泳总路程显著增加(P<0.01)。穿越平台次数,目标象限停留时间及路程显著减少(P<0.01),大鼠海马CA3区神经元细胞数量减少排列紊乱,神经元树突分支数量及单位长度树突棘密度显著下降(P<0.01),海马CA3区PSD95、SYN1、GAP43及RAC1蛋白表达水平显著降低(P<0.01);与M组相比,ME组和MS组平台潜伏期和游泳总路程有所降低(P<0.05),穿越平台次数,目标象限停留时间及路程有所增加(P<0.05),大鼠海马CA3区神经元细胞数量增加排列整齐,神经元树突分支数量及单位长度树突棘密度有所增加(P<0.05),海马CA3区PSD95、SYN1、GAP43及RAC1蛋白表达水平增加(P<0.05),而MES组平台潜伏期和游泳总路程明显降低(P<0.01),穿越平台次数,目标象限停留时间及路程明显增加(P<0.01),大鼠海马CA3区神经元细胞数量增加排列整齐,神经元树突分支数量及单位长度树突棘密度明显增加(P<0.01),海马CA3区PSD95、SYN1、GAP43及RAC1蛋白表达水平显著增加(P<0.01)。结论 运动结合生慧汤可以协同增效的防止睡眠剥夺大鼠学习记忆损伤,及其作用机制可能是通过上调SYN、PSD95、GAP43及RAC1蛋白来保护神经元树突分� Objective To investigate whether exercise combined with Shenghui decoction can synergistically prevent learning memory impairment in sleep deprived rats,and its possible mechanism of action.Methods Fifty SD rats were randomly divided into blank group(group C),model group(group M),model+exercise group(group ME),model+shenghui decoction group(group MS) and model+exercise+shenghui decoction group(group MES),and the ME group was subjected to 4 weeks of running table exercise,the MS group was subjected to 4 weeks of gavage with raw wei tang,while the MES group was subjected to 4 weeks of both running table exercise and gavage with raw wei tang,followed by sleep deprivation using a multi.The MES group was subjected to 72 h of sleep deprivation using a multi-platform water environment.After the sleep deprivation,the learning memory ability of each group was examined by Morris water maze,and the changes of neuronal morphology,dendritic branching spine unit dendritic spine density in hippocampal area were observed by HE staining and Golgi staining,and the expression levels of PSD95,SYN1,GAP43 and RAC1 in CA3area of hippocampus were detected by immunoprotein blotting.Results Compared with group C,platform latency and total swimming distance were significantly increased in group M rats(P0.01).The number of platform crossing,target quadrant dwell time and distance traveled were significantly reduced(P0.01),the number of neuronal cells in CA3 area of hippocampus was reduced and disorganized,the number of neuronal dendritic branches and dendritic spine density per unit length were significantly reduced(P0.01),and the expression levels of PSD95,SYN1,GAP43 and RAC1 in CA3 area of hippocampus were significantly reduced(P0.01).Compared with the M group,the ME and MS groups showed a decrease in platform latency and total swimming distance(P0.05),an increase in the number of platform crossings,target quadrant dwell time and distance(P0.05),an increase in the number of neuronal cells in the CA3region of the hippocampus with neural al
