摘要
目的分析成纤维细胞生长因子-23(FGF-23)基因多态性与乙肝病毒(HBV)感染孕妇应用富马酸丙酚替诺福韦(TAF)阻断效果的相关性,提高HBV感染孕妇治疗效果。方法选取2017年5月-2020年5月南阳市第二人民医院收治的96例HBV感染孕妇作为研究对象,按照TAF母婴传播免疫阻断结果将孕妇分为阻断成功组64例与阻断失败组32例,采用限制性片段长度多态性聚合酶链反应(PCR-RFLP)检测FGF-23基因多态性,采用Logistic回归分析FGF-23基因多态性与HBV感染孕妇TAF阻断效果的关系。结果阻断成功组FGF-23基因rs7955866位点GG基因频率及G等位基因频率和rs3812822位点TT基因频率及T等位基因频率均高于阻断失败组(P<0.05),两组rs13312756位点基因频率及等位基因频率比较差异无统计学意义。Logistic回归分析显示,FGF-23基因rs7955866位点AA基因型和rs3812822位点CC基因型可能增加HBV感染孕妇TAF阻断失败风险(P<0.05)。结论FGF-23基因rs7955866位点AA基因型和rs3812822位点CC基因型与HBV感染孕妇TAF阻断效果有关。
OBJECTIVE To analyze the association of fibroblast growth factor-23(FGF-23)gene polymorphism with blocking effect of tenofovir alafenamide fumarate(TAF)on hepatitis B virus(HBV)infection in pregnant women so as to improve the curative effect of the pregnant women with HBV infection.METHODS A total of 96 pregnant women with HBV infection who were treated in Nanyang Second People′s Hospital from May 2017 to May 2020 were recruited as the research subjects and were divided into the successful blocking group with 64 cases and the failed blocking group with 32 cases according to the immunologic blocking outcomes of TAF on mother-to-infant transmission.The FGF-23 gene polymorphism was detected by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP),and logistic regression analysis was performed for the correlation between the FGF-23 gene polymorphism and the blocking effect of TAF on HBV infection.RESULTS The frequencies of GG genotype and G allele at rs7955866 locus of FGF-23 gene and TT genotype and T allele at rs3812822 locus were higher in the successful blocking group than in the failed blocking group(P<0.05).There were no significant difference in the frequencies of genotypes and alleles at rs13312756 locus between the two groups.Logistic regression analysis showed that the AA genotype at rs7955866 locus and CC genotype at rs3812822 locus of FGF-23 gene may increase the risk of failed blocking of TAF on HBV infection(P<0.05).CONCLUSION The AA genotype at rs7955866 locus and CC genotype at rs3812822 locus of FGF-23 gene are associated with the blocking effect of TAF on HBV infection in the pregnant women.
