摘要
目的探究硒化甘草多糖(SeGUP)和甘草多糖(GUP)对产志贺毒性大肠杆菌(STEC)的体外抑菌机制和抗氧化效果。方法采用二倍稀释法检测SeGUP、GUP对STEC的半数抑制浓度(IC_(50)),测定各浓度的SeGUP、GUP对STEC生长曲线的影响;通过测定不同时间段培养液中AKP、β-半乳糖苷酶(β-gal)、蛋白质含量以及结晶紫染色探究SeGUP、GUP对STEC细胞壁、细胞膜、生物膜的影响。采用ELISA法检测清洁级昆明小鼠血清中SOD、CAT、MDA含量变化。结果SeGUP IC_(50)为55.58 mg·mL^(-1),GUP IC_(50)为69.85 mg·mL^(-1),150 mg·mL^(-1)SeGUP能完全抑制STEC生长,GUP在200 mg·mL^(-1)才能完全抑制STEC的增殖。SeGUP组同一时间的AKP活性、β-gal活性、蛋白质含量与GUP组比较,差异极显著,P<0.01;SeGUP组与GUP组比较能极显著降低STEC生物膜形成,P<0.01。抗氧化试验中,SeGUP组提高SOD、CAT含量以及降低MDA含量效果优于GUP组(P<0.01)。结论硒化修饰提高了GUP体外抑菌效果、破坏STEC膜结构和抗氧化能力。
Objective The study aimes to investigate the bacteriostatic mechanism of selenide Glycyrrhiza polysaccharide(SeGUP)and Glycyrrhiza polysaccharide(GUP)against Shiga toxin-producing E.coli(STEC)in vitro,and to analyze the antioxidant effects of SeGUP and GUP.Methods In vitro bacteriostatic test,the half-inhibitory concentration(IC_(50))of SeGUP and GUP on STEC was detected by two-fold dilution method.Determination of the effect of each concentration of SeGUP and GUP on the growth curve of STEC.The effects of SeGUP and GUP on the cell wall,cell membrane and biofilm damage of STEC were investigated by in culture medium at different time periods measuring AKP,β-galactosidase(β-gal),protein contents and crystalline violet staining.The changes of SOD,CAT and MDA contents in the serum of clean grade Kunming mice were detected by ELISA.Results SeGUP IC_(50)of 55.58 mg·mL^(-1)and GUP IC_(50)of 69.85 mg·mL^(-1).150 mg·mL^(-1)SeGUP completely inhibited STEC growth,while 200 mg·mL^(-1)GUP could completely inhibit the proliferation of STEC.The AKP activity,β-gal activity and protein contents of SeGUP group were extremely significant different from those of GUP group at the same time(P<0.01);Compared with GUP group,SeGUP group could extremely significant reduce the formation of STEC biofilm,P<0.01;Antioxidant test results showed that SeGUP group was better than GUP group in increasing SOD,CAT and reducing MDA contents(P<0.01).Conclusion The selenium modification improved the bacteriostatic effect of in vitro,destroyed the structure of membrane on STEC and antioxidant capacity of GUP.
