艾灸对癌性疲劳小鼠免疫神经内分泌的影响 被引量:2

Effects of moxibustion on immuno-neuro-endocrine in mice with cancer related fatigue

摘要 观察艾灸对癌性疲劳(CRF)模型小鼠疲劳状态及免疫神经内分泌的影响。方法:将BALB/C小鼠随机分为空白组10只、模型组15只、艾灸组15只、红外组15只。选择乳腺癌细胞系皮下移植联合顺铂构建CRF小鼠模型,各治疗组干预关元穴4周后进行小鼠行为学观察(疲劳跑台实验、悬尾实验),并运用ELISA检测促肾上腺激素释放素(CRH)、促肾上腺皮质激素(ACTH)、皮质醇(CORT)、白细胞介素(IL)-1β、IL-6、IL-10含量,Western Blot法检测蛋白激酶B(AKT)、p-AKT、核因子κB p65(NF-κB p65)、p-NF-κB p65蛋白表达,免疫荧光法检测下丘脑CRH、脾脏IL-6表达。结果:与空白组比较,模型组小鼠疲劳奔跑时间及距离显著缩短(P<0.01),CORT、IL-10含量显著减少(P<0.01),跑台电击次数及悬尾不动时间显著增加(P<0.01),CRH、ACTH、IL-1β、IL-6含量显著升高(P<0.01),下丘脑CRH及脾脏IL-6荧光表达显著增加(P<0.01)。与模型组比较,各干预组小鼠奔跑时间及距离显著增加(P<0.01,P<0.05),IL-10含量显著升高(P<0.01),跑台电击次数和悬尾不动时间显著减少(P<0.05,P<0.01),CRH、ACTH、IL-6含量显著降低(P<0.01,P<0.05),下丘脑CRH及脾脏IL-6荧光表达显著减少(P<0.05),p-AKT/AKT、p-NF-κB p65/NF-κB p65蛋白表达显著减少(P<0.05,P<0.01)。与红外组比较,艾灸组悬尾不动时间缩短(P<0.05),ACTH、IL-6含量显著降低(P<0.01,P<0.05),p-NF-κB p65/NF-κB p65蛋白表达显著减少(P<0.01)。结论:艾灸可改善CRF症状,其中红外光发挥了重要作用,其机制可能通过调节AKT-NF-κB炎症通路及下丘脑-垂体-肾上腺轴发挥作用。 Objective:To observe the effects of moxibustion on fatigue status and immune neuroendocrine in cancer related fatigue(CRF)model mice.Methods:BALB/C mice were randomly divided into the blank group(n=10),the model group(n=15),the moxibustion group(n=15),and the infrared group(n=15).The subcutaneous transplantation of breast cancer cell lines combined with cisplatin was selected to construct a CRF mouse model,and behavioral observations(fatigue running table test and tail suspension test)were performed in each treatment group after 4 weeks of intervention at Guanyuan acupoint,and ELISA was applied to detect adrenal hormone releasing hormone(CRH),adrenocorticotropic hormone(ACTH),cortisol(CORT),interleukin(IL)-1β,IL-6,and IL-10 content,protein kinase B(AKT),p-AKT,nuclear factor kB p65(NF-B p65),p-NF-kB p65 protein expression by Western Blot,and hypothalamic CRH and splenic IL-6 expression by immunofluorescence.Results:Compared with the blank group,the mice in the model group had significantly shorter fatigue running time and distance(P<0.01),significantly reduced CORT and IL-10 content(P<0.01),significantly increased number of electric shocks and hanging tail immobility time on the running platform(P<0.01),significantly increased CRH,ACTH,IL-1β,IL-6 content(P<0.01),hypothalamic CRH and splenic IL-6 fluorescence expression were significantly increased(P<0.01).Compared with the model group,the mice in each intervention group had increased running time and distance(P<0.01,P<0.05),increased IL-10 content(P<0.01),decreased number of electric shocks and hanging tail immobility time on the running platform(P<0.05,P<0.01),decreased CRH,ACTH,IL-6 content(P<0.01,P<0.05),hypothalamic CRH and splenic IL-6 fluorescence expression was reduced(P<0.05),and p-AKT/AKT and p-NF-B p65/NF-kB p65 were reduced(P<0.05,P<0.01).Compared with the IR group,the mice in moxibustion group had shorter hanging tail immobility time(P<0.05)and reduced ACTH,IL-6 content(P<0.01,P<0.05),and reduced the protein expression level of p-NF-kB p65(P<0.01).Co
