前列腺癌中ZIC2与驱动蛋白KIF18B表达及相互关系与预后风险评估

Expression of ZIC2 and kinesin KIF18B in prostate cancer and their relationship with prognosis risk assessment

摘要 目的检测前列腺癌中ZIC2、KIF18B表达水平和二者相关性及与预后风险的评估价值。方法收集根治性切除的前列腺癌组织标本32例及同期良性前列腺增生(BPH)组织36例,免疫组化SP法检测ZIC2及KIF18B的表达,分析其表达变化相关性及与前列腺特异抗原(PSA)、Gleason评分[国际泌尿病理(ISUP)分级]和T分期等预后风险因素的关系。结果ZIC2在PCa组织中和BPH中的阳性表达率分别为87.5%(28/32)及19.4%(7/36),有统计学差异(χ^(2)=28.976,P=0.000);前列腺癌及BPH中KIF18B阳性表达率分别为84.4%(27/32)及38.9%(14/36),有统计学差异(χ^(2)=14.641,P=0.000);ZIC2表达与T分期密切相关(χ^(2)=15.013,P=0.000);KIF18B表达与Gleason评分(ISUP分级)存在相关性(χ^(2)=5.709,P=0.038)。结论前列腺癌中ZIC2、KIF18B表达异常升高,均属癌基因;二者异常表达分别与T分期及Gleason(ISUP分级)相关,同时检测有助于前列腺癌预后的临床判定。 Objective To detect the expression levels of ZIC2 and KIF18B in prostate cancer(PCa)and explore their correlation,as well as their value in evaluating prognostic risk.Methods 32 patients with radical resection of PCa and 36 patients with benign prostatic hyperplasia(BPH)at the same time were collected,the expression levels of ZIC2 and KIF18B were detected by immunohistochemical SP method.The correlation of their expression changes and the relationships between prognostic risk factors such as prostate-specific antigen(PSA),Gleason score[International Society of Urological Pathology(ISUP)grade]and T stages were analyzed.Results The positive expression rates of ZIC2 in PCa tissue and BPH were 87.5%(28/32)and 19.4%(7/36),respectively,and there was a significant difference between them(χ^(2)=28.976,P=0.000).The positive expression rates of KIF18B in PCa and BPH were 84.4%(27/32)and 38.9%(14/36),respectively,and there was a significant difference between them(χ^(2)=14.641,P=0.000).The expression level of ZIC2 was closely correlated with T stages(χ^(2)=15.013,P=0.000).The expression of KIF18B was correlated with Gleason score(ISUP grade)(χ^(2)=5.709,P=0.038).Conclusions The expressions of ZIC2 and KIF18B are abnormally elevated in PCa,both of which belong to oncogenes.Abnormal expressions of the two genes are correlated with T stages and Gleason(ISUP grade),respectively,and simultaneous detection both are helpful for clinical determination of prognosis of PCa.
