不同规格的日本鳗鲡肠道菌群结构与功能的差异分析

Analysis of the structure and functional differences of intestinal flora in Anguilla japonica of different sizes

摘要 为了揭示不同生长速率日本鳗鲡的肠道菌群结构,为阐明肠道菌群对日本鳗鲡生长速率的影响提供参考依据,利用16S rDNA测序技术对同一养殖条件下不同规格的日本鳗鲡前肠、中肠和后肠的菌群结构和功能进行分析,结果显示不同规格日本鳗鲡肠道微生物门和属水平的菌群结构差异明显。小规格组鳗鲡肠道内的OTU数目、Chao、Shannon指数均高于大规格组;软壁菌门(Tenericutes)、厚壁菌门(Firmicutes)和变形菌门(Proteobacteria)为日本鳗鲡肠道内优势菌门且在两组间均无显著性差异。大规格组前肠中支原菌属(Mycoplasma)、鲸蜡杆属(Cetobacterium)、细小杆菌属(Sediminibacterium)的丰度显著高于小规格组(P<0.05),而梭杆菌门(Fusobacteria)、变形菌门(Proteobacteria)和拟杆菌门(Bacteroidetes)的丰度显著低于小规格组(P<0.05);中肠中大规格组支原菌属(Mycoplasma)的丰度显著高于小规格组(P<0.05),而单胞菌属(Plesiomonas)、梭菌属(Clostridium)、罗尔斯通菌属(Ralstonia)的丰度显著低于小规格组(P<0.05);后肠中大规格组Candidatus、支原菌属(Mycoplasma)的丰度显著高于小规格组,而鲸蜡杆属(Cetobacterium)、单胞菌属(Plesiomonas)的丰度显著低于小规格组(P<0.05)。在大规格组中与环境适应、信号转导、跨膜运输、免疫、基因信息处理等功能相关的微生物的丰度高于小规格组(P<0.05),小规格组中与氨基酸代谢、脂代谢、外源物质代谢等代谢功能相关菌群的丰度较高。研究结果表明不同规格日本鳗鲡的肠道菌群结构存在显著性差异,有助于揭示造成养殖日本鳗鲡生长速率差异的原因。 In order to reveal the structure of the intestinal microflora of Anguilla japonica with different growth rates and provide a reference for clarifying the effect of the intestinal microflora on the growth rate of A.japonica,the microflora structure and function of the foregut,midgut and hindgut of different sizes A.japonica under the same cultured conditions were analyzed by 16s rDNA sequencing technology.The results showed that there were significant differences in the structure of gut flora between different sizes Anguilla japonica at the level of genus and phylum.The number of OTU,Chao and Shannon index in the intestine of small eels were higher than those of the big eels;Tenericutes,Firmicutes and Proteobacteria were the dominant phyla in the intestine and there was no significant difference between the two groups.The abundance of Mycoplasma,Cetobacterium,and Sediminibacterium in the foregut of big Anguilla japonica were significantly higher than those in the small eels(P<0.05),while the abundance of Fusobacteria,Proteobacteria,and Bacteroidetes were significantly lower than those of the small eels(P<0.05).The abundance of Mycoplasma in the midgut of big Anguilla japonica was significantly higher than that in the small eels(P<0.05),while the abundance of Plesiomonas,Clostridium,and Ralstonia were significantly lower than those in the small eels(P<0.05).The abundance of Candidatus and Mycoplasma in the hindgut of big Anguilla japonica were significantly higher than those in the small eels,while the abundance of Cetobacterium and Plesiomonas were significantly lower than those in the small eels(P<0.05).In the intestine of big A.japonica,the abundance of microorganisms associated with signal transduction,membrane transport,environmental adaptation,immunity,and gene information processing were higher than those in the small eels(P<0.05),while the abundance of microorganisms associated with lipid metabolism,amino acid metabolism,exogenous material metabolism were higher than those in the small eels.The results of t
