青海省果洛州、黄南州鼠疫菌CRISPR基因分型及其多态性分析

CRISPR genotyping and polymorphism analysis of Yersinia pestis in Guoluo and Huangnan Prefecture of Qinghai Province

摘要 目的了解青海省果洛、黄南州鼠疫菌CRISPR遗传多态性和地区分布特征,为该地区鼠疫防控工作提供科学依据。方法采用常规方法对实验菌株进行DNA提取,利用PCR技术对设计的相应引物的CRISPR 3个位点YPa、YPb、YPc进行扩增、测序,通过软件工具CRISPR Finder和NCBI数据库,分析鉴定其间区序列,推测基因序列来源。结果26株鼠疫菌在CRISPR 3个位点发现17种间区序列,分别为YPa 9种、YPb 5种、YPc 3种。所有试验用菌株可分为5种CRISPR基因型(G7、G22、G22-a1′、G24、G30),其中G22和G22-a1′为该地区主要基因型。结论青海省果洛、黄南州鼠疫菌CRISPR基因遗传位点具有多态性,地理分布区域特征明显,为本地区建立鼠疫菌基因库提供基础数据。 Objective To understand the CRISPR genetic polymorphism and regional distribution characteristics of Yersinia pestis in Guoluo and Huangnan Prefecture in Qinghai Province,to provide a scientific basis for plague prevention and control in the region.Methods Conventional methods were used to extract DNA from the experimental strains.PCR(polymerase chain reaction)technology was used to amplify and sequence the three CRISPR sites YPa,YPb and YPc with the corresponding primers.The software tools CRISPR Finder and NCBI database were used to analyze and identify the inter-regional sequences,and infer the gene sequence origin.Results Twenty-six strains of Y.pestis were identified 17 inter-regional sequences at three CRISPR sites,namely 9 species of YPa,5 species of YPb,and 3 species of YPc.All tested strains could be classified into five CRISPR genotypes(G7,G22,G22-a1′,G24 and G30),among which G22 and G22-a1′were the main genotypes in the region.Conclusion The genetic loci of the CRISPR gene of Y.pestis in Guoluo and Huangnan Prefecture of Qinghai Province are polymorphic and had obvious geographical distribution characteristics.The study provided basic data for the establishment of the gene bank for Y.pestis in the region.
