表皮生长因子受体靶向性纳米探针的肺癌细胞靶向性能与双光子荧光成像

Epidermal growth factor receptor targeted fluorescent probes for lung cancer cells and their two-photon fluorescence imaging

摘要 目的探讨新型具有聚集诱导发光性质的纳米探针(AIE-225)对表皮生长因子(EGFR)的靶向结合能力,细胞器靶向性及其双光子成像效果。方法采用HCC827,HLF细胞作为模型细胞,探讨其细胞结合能力,以商业溶酶体染料为参比对象,探讨其亚细胞水平的靶向区域和靶向能力,同时应用多种波长的双光子激光(如850、950 nm)进行激发,探讨该纳米探针的双光子靶向成像能力。结果AIE-C225探针具有良好的靶向性,对EGFR高表达的HCC827细胞具有良好的靶向性能,与之比较,使用未耦联西妥昔单抗的AIE纳米颗粒未见到荧光,同时采用正常肺永生化细胞HLF也没荧光信号。AIE-C225作用的HCC827细胞其细胞器定位为溶酶体,与商用溶酶体染料具有高达0.88的共定位系数,且其在850 nm和950 nm的双光子激发下的的细胞荧光成像与488 nm激发下的共定位系数分别为0.991、0.998(Manders系数)。结论AIE-C225具有良好的肺癌细胞EGFR靶向性能,其良好的双光子吸收、荧光发射性能为其后续双光子光动力研究提供基础技术支持。 Objective To investigate the target binding ability of the nanoprobe(AIE-225),which is endowed with aggregation-induced emission(AIE)property,on epidermal growth factor(EGFR),its organelle targeting ability,and the corresponding two-photon imaging abilities.Methods HCC827 and HLF cells were chosen as model cells to explore their cell binding ability,and commercial lysosomal dyes were used as a reference to explore its targeting subcellular regions and targeting ability at the subcellular level,while various two-photon lasers(e.g.850 nm,950 nm)were applied for two-photon excitation to explore the two-photon targeting imaging ability of AIE-225 nanoprobe.Results The AIE-C225 probe had good targeting performance to HCC827 cells with high EGFR expression.In comparison,no fluorescence was seen with AIE nanoparticles that were not coupled with cetuximab(C225),and no fluorescence signal was seen when using normal lung immortalized cells(HLF)either.AIE-225 probe was performed with excellent targeting performance towards the subcellular organelles lysosome and it had a high colocalization coefficient of 0.88 with the commercial lysosomal probe,LysoTracker™Deep Red.The two-photon imaging ability of AIE-225 under two-photon excitation at 850 nm and 950 nm was excellent.Its co-localization coefficients with the images of 488 nm excitation were 0.991 and 0.998 under two-photon excitation at 850 nm and 950 nm,respectively.Conclusion AIE-C225 possesses good targeting performance in lung cancer cells which overexpressed EGFR.Its good two-photon absorption and fluorescence emission performance provide basic technical support for its subsequent two-photon photodynamic studies.
