水蛭素对糖尿病肾病大鼠肾损伤保护作用研究

摘要 目的:观察水蛭素(hirudin)对大鼠糖尿病肾病模型肾损伤的保护作用。方法:雄性SD大鼠70只随机分为空白对照组(K组,10只),造模组(60只)。造模组用高糖高脂饮食联合注射链脲佐菌素(STZ)诱导糖尿病肾病(DN)模型并随机分为DN模型组(M组)、水蛭素低、中、高(D、Z、H)组、贝那普利组(B组),每组10只,灌胃给药15周。观察各组大鼠一般状态,干预结束收集大鼠24h尿液、血液,检测大鼠血糖浓度、糖化血红蛋白(HbA1c),血清尿素氮(BUN)、血肌酐(SCR),尿肌酐(UCr)、尿β_(2)-微球蛋白(β_(2)-MG)等指标。结果:与K组比较M组大鼠各项指标显著增高(P<0.01)。与M组比较Z组大鼠血糖较低(P<0.05),HbA1c、BUN、SCR、cys C、24h UV、β_(2)-MG较低(P<0.01),UCr则无明显差异(P>0.05),H组大鼠各项指标更低(P<0.01)。结论:水蛭素可减轻DN模型大鼠肾损伤,延缓DN发展进程,具有保护肾功能的作用,并且存在明显的剂量效应关系。 Objective:To observe the protective effect of hirudin on renal injury in rats with diabetic nephropathy.Methods:Seventy male SD rats were randomly divided into blank control group(K group,n=10)and model group(n=60).The diabetic nephropathy(DN)model was induced by injection of streptozotocin(STZ)with high-sugar and high-fat diet and randomly divided into DN model group(M group),low,medium and high(D,Z,H)dosage of hirudin groups,Benazepril group(B group),10 rats in each group,intragastric administration for 15 weeks.The general state of rats in each group was observed.At the end of intervention,24 h urine and blood of rats were collected to detect blood glucose concentration,glycosylated hemoglobin(HbA1c),serum urea nitrogen(BUN),serum creatinine(SCR),urinary creatinine(UCr),urinaryβ2-microglobulin(β2-MG)and other indicators.Results:Compared with group K,the indexes in group M were significantly increased(P<0.01).Compared with group M,the blood glucose in group Z was lower(P<0.05),HbA1 c,BUN,SCR,cys C,24 h UV andβ2-MG were lower(P<0.01),UCr was not significantly different(P>0.05),and the indexes in group H were lower(P<0.01).Conclusion:Hirudin can reduce renal injury in DN model rats,delay the development of DN,protect renal function,and there is a significant dose-effect relationship.
